材料和方法
1B8拮抗剂分子的构建
将该分子构建成使得GH分子(G120R)的位点2(低亲和力位点)中的氨基酸甘氨酸-120突变成精氨酸。GH分子通过高亲和力位点1与GH受体的结合不受影响,然而,通过GH位点2与受体结合受到精氨酸分子的大侧基抑制。
预先将PCR策略用于产生含有G120R突变的GH分子,并且使用合适的限制酶切位点,使得该分子克隆至pTrc-His表达质粒中,以产生克隆pTrc-His-1A7(G120R与GHR细胞外B结构域连接)。
然后从该载体切除300bp Bsu36I-Not1片段,并将其连接至哺乳动物表达质粒pGHsecTag-1B7中(GH与GHR细胞外结构域A和B连接),以产生pGHsecTag-1B8(通过GH分泌信号指导分泌性表达)。参见图9。
1B9v0拮抗剂分子的构建
已经将该分子构建成使得GH分子的位点2和位点2二者中的氨基酸均突变。这些突变增强了GH分子通过高亲和力位点1与GH受体的 结合,而单甘氨酸至精氨酸的改变抑制通过GH位点2与受体的结合。

1B8v0和1B9v0的变体克隆的构建

拮抗剂变体分子的体外生物活性
使用GH特异性荧光酶报告子测定法检测各嵌合体的体外生物活性。基本上,人源细胞系被人GH受体稳定转染,且然后被荧光酶信号传导报告子瞬时转染。该测定法检测GH的生理学水平,见图14。
拮抗剂分子的纯化
在无蛋白培养基中培养CHO Flp-In细胞系,该细胞系表达1B8v2和1B9v2作为分泌性产物。亲和纯化之前收获、浓缩以及澄清培养基。对于纯化,制备20ml与抗hGH的5E1单克隆抗体偶联的NHS激活的Sepharose 4 Fast Flow树脂。在纯化之前,通常10倍浓缩培养基样品,并使用结合缓冲液(25mM Tris HCl/150mM NaCl,pH 7.4)1∶1稀释。
将材料以2ml/min流速上样于的柱子上。洗涤之后,使用200mM 的甘氨酸(pH 2.7)以1ml/min洗脱结合蛋白,然后使用1M Tris HCl,pH9.0中和。通过SDS-PAGE(见图15a和图15b)分析样品。图17a和图17b描述了与非变性生长激素嵌合体相比较的1B9的二聚体形成。
1B8的药物代谢动力学研究
皮下(SC,图16a)或静脉内(IV,图16b)给予6只正常健康大鼠单剂量注射的1nMol(75μg)蛋白。对照大鼠仅给予媒介(vehicle)。在10天期间按照时间间隔取样,并使用自建的GH酶联免疫吸附测定来分析1B8的存在。
