技术领域
本发明属生物技术领域,本专利主要涉及PSA2在制备蛋白与基因诊断试剂盒中的应用。可用于前列腺癌的早期诊断及治疗效果的监测。
背景技术
自从,前列腺特异性抗原(prostate specific antigen,PSA)用于人群筛查以来,前列腺癌的诊断发生了划时代的变化。美国早在1983年首先应用PSA进行人群前列腺癌筛查,历时20年实现了早期诊治。然而,只具备前列腺组织特异性的PSA尚存在如下缺点,①集团普查中PSA>4.0ng/ml的人群中只有25-30%的人诊断为前列腺癌,无效活检率高达70%;②PSA对前列腺腺癌以外的特殊类型癌,如印戒细胞癌鳞样细胞癌等无诊断价值,漏诊的病例数约占癌病例总数的8%;③难以鉴别早期前列腺癌与良性前列腺增生所引起的PSA轻度增高。因此,探索新的前列腺癌标志蛋白成为亟待解决难题。
PSA是一种前列腺上皮细胞产生并分泌于血清中的丝氨酸蛋白酶,具有很高的前列腺组织特异性。PSA在血液中以三种形式存在:即自由分子形式(FPSA)、与α12抗糜蛋白酶的复合物形式(PSA-ACT)及与α22巨球蛋白酶的复合物形式(PSA-α2M),其中只有极少部分以FPSA形式存在。正常情况下,前列腺上皮细胞分泌的PSA进入前列腺腺腔内,随前列腺液排除,并成为精液的重要成份之一。由于,前列腺上皮细胞周围有基底细胞层构成屏障,正常时PSA只通过管腔侧排除,血清PSA含量低于4.0ng/ml。前列腺癌时,癌性腺体缺乏基底细胞层,分泌的PSA直接进入前列腺间质中经毛细血管入血而使血清PSA含量增高。尽管PSA在前列腺癌早期诊断中有上述缺点,但是,迄今尚无可以取代它的标志蛋白。
PSA2的发现
PSA2是应用SELDI-TOF蛋白质芯片技术,通过血清差异蛋白质谱学研究发现的一个分子量为15868的前列腺癌高表达蛋白。
血清PSA2蛋白的分离 纯化与鉴定
分离纯化与质谱鉴定:取高表达PSA2蛋白的血清样本,IMAC-CU柱纯化获得蛋白浓缩液,SDS-PAGE电泳,切取相应分子量的蛋白带,MALDI-TOF鉴定,提供了类似的蛋白信息(北京华大基因有限公司);
15868蛋白的N-末端测序:应用美国ABI公司的PROCISE491测序仪,进行了该标志蛋白N末端测序(中国医学科学院基础医学研究所,中心实验室完成),依据测序结果进行数据库查询,证明PSA2是LZTR2蛋白中的一个片段。因人LZTR2(Leucine Zipper Transcription Regulator 2)的15868片段在前列腺癌诊断中有与PSA互补作用,因此,命名为PSA2。LZTR2(Leucine Zipper TranscriptionRegulator 2),又称为RGPR(Reducacion Gene Promotor Region Related Protein)。
发明内容
本发明提供一种人重组多肽PSA2在制备检测前列腺癌试剂盒中的应用,以解决目前PSA诊断试剂盒的诊断特异性较低的问题。本发明通过检测血中PSA2多肽含量,与血清PSA值相配合,提高前列腺癌的诊断效率。
PSA2多肽与PSA诊断的一致率为72.3%,诊断的互补作用极高。在83例前列腺癌患者中仅5例为PSA<4.0ng/ml,而PSA2则明显增高可准确地诊断出PSA漏诊的病例;在其余78例PSA含量>4.0ng/ml的前列腺癌患者中,PSA2信躁比低于界定值5者为17例,均被PSA准确诊断出来;在83例前列腺癌患者中PSA和PSA2均增高者为61例,诊断一致率为72.3%,没有1例被二者漏诊的病例。因其在前列腺癌诊断中有与PSA高度互补作用,命名为PSA2。

本发明所述的人重组多肽PSA2抗原是指人重组多肽PSA2纯蛋白抗原;所述的免疫动物包括各种免疫动物如马、绵羊、兔子、豚鼠等。
本发明人重组多肽PSA2诊断试剂盒中采用的人重组多肽PSA2抗体是人重组多肽PSA2蛋白抗体。该抗体包括单克隆抗体和多克隆抗体。
多肽PSA2蛋白抗体,该抗体为基础衍生出的人源化抗体在制备前列腺癌治疗药物中的应用。
一种前列腺癌特异性蛋白PSA2的检测试剂盒,包括包被有抗体的酶标板、酶标二抗、通用试剂Tween-20和显色底物四甲基联苯胺TMB,其特征在于该试剂盒的酶标板孔内为经包被液处理和阻滞液处理的PSA2融合蛋白抗体,另配备有辣根过氧化物酶HRP标记的兔抗PSA2多肽抗体。

本发明公开的人重组多肽PSA2的制备方法如下:
采用组氨酸标签融合蛋白表达人多肽PSA2:
采用多聚酶链式反应(PCR)方法克隆人重组多肽PSA2基因蛋白编码序列,构建该基因的融合表达质粒,导入BL2 1的大肠杆菌中,I PTG诱导其在大肠杆菌中的表达,SDS-PAGE电泳,western blot鉴定,用组氨酸标签融合蛋白纯化试剂盒纯化融合蛋白,用HPLC大量分离与纯化蛋白质,以制备兔多抗和小鼠单抗,制备用于前列腺癌早期诊断的人重组多肽PSA2基因诊断试剂盒。具体如下:
据原核表达载体的组氨酸标签阅读框架设计引物,使人重组多肽PSA2基因的编码区阅读框与组氨酸标签的阅读框架一致。以含PSA2基因的质粒为模板(O.1ug),加入上述引物各10pmol进行扩增,回收PCR产物条带。用Sma 1 I酶切PCR回收产物及表达载体,产生匹配的粘末端。小胶回收试剂回收线性的4.9kb的载体,PSA2编码区片段。将表达载体与人重组多肽PSA2基因连接,构建成符合读框的组氨酸标签PSA2融合基因。用含融合基因的重组质粒转化大肠杆菌BL2 1,小量制备质粒DNA,Sa l I和Not I酶切鉴定是否含插入片段,含插入片段的阳性克隆质粒DNA纯化后用DNA自动测序仪测序。设计合成重组人多肽PSA2引物序列:

单个阳性菌落接种入含氨卞青霉素(100ug/ml)的2ml LB培养液中,37℃,过夜培养。将400ul过夜培养物分别接种到40ml含氨卞青霉素(1OOug/ml)的LB培基中,于37℃培养7h,加入IPTG至终浓度为1mmol/L,37℃,培养2.5h,转移至50ml离心管中。室温12000×g离心10min,去尽上清,置于冰上,重悬于预冷的600ul的PBS中。加入终浓度为100ug/ml的溶菌酶、10ug/ml DNa seI,用液氮和温水介导的反复冻融法破膜,11000rp离心10min,收集上清。重悬组氨酸标签MicroSpin纯化柱中的树脂,打开盖及底盖,置于一干净的1.5ml的离心管中,735×g离心1min,弃液体,盖上底盖,柱中加入600ul的细菌裂解液。盖紧上盖,轻轻混匀,置于室温5-10min后打开盖及底盖,置于一干净的1.5ml的离心管中,735×g离心1min,再用400ul PBS用相同的方法洗涤2次。柱中加入200ul的谷胱苷肽洗脱液,盖紧上盖,轻轻混匀,置于室温5~10min。735×g离心1min收集流出液,获得人多肽PSA2(见下图)。
本发明公开的人多肽PSA2诊断试剂盒的制备包括下列步骤:
1、抗体制备
取健康免疫动物,用人多肽PSA2抗原进行免疫,第一次以抗原加卡介苗和福氏佐剂研匀,注射动物颈淋巴结或皮下肌肉组织,每二周一次,共免疫4-5次,抗原量抗原量根据动物体重增减,免疫最后一次的二周后采血,动物抗人血清用饱和硫酸铵二级沉淀后上蛋白A/G亲和层析,得到动物抗人人重组多肽PSA2 IgG抗体,测定效价,冷冻备用;
制备兔抗人多肽PSA2抗原的合成多肽抗体:
①、根据的氨基酸序列,设计并化学合成21个氨基酸的PSA2多肽,该多肽的序列为:YYGRKKATLEWAMKNHLWGHC,将多肽C端与钥孔戚血蓝素(keyhole limpet hemocyanin,KLH)交联以增加免疫原性。
②、将多肽溶于磷酸盐(PBS)缓冲液,注射至新西兰家兔,首次剂量为300ug/kg,加强免疫的剂量约为首次剂量的1/4左右。每2~3周加强免疫一次,共免疫4次,获得的抗血清用Pierce公司的亲和层析纯化试剂盒(具体步骤严格按Pierce公司的说明书进行)纯化出抗PSA2的IgG;
2、抗体标记
取人重组多肽PSA2 IgG抗体采用常规过碘酸钠法标记辣根过氧化物酶;
3、包被抗体聚苯乙烯微孔板制备
取标记的人重组多肽PSA2 IgG抗体,工作浓度于50mM,pH9.5碳酸盐条件下加入聚苯乙烯微孔,每孔100微升,并于2-8℃条件放置约40小时;取出用生理盐水洗涤5次,烘干,用铝箔袋密封包装。
4,其它应用试剂:
(1)包被液:0.05molpH9.5碳酸盐溶液;
(2)洗涤液:0.8%氯化钠--0.05%Tween20;
(3)稀释液:0.15molpH7.2磷酸盐一氯化钠溶液;
(4)显色缓冲液:0.05molpH5.0磷酸盐一柠檬酸溶液;
(5)显色剂;邻苯二铵;
(6)终止液:2M硫酸溶液。
人重组多肽PSA2诊断试剂盒具体制备方法:
每次实验每块包被抗体聚苯乙烯微孔板设人重组多肽PSA2参考标准品(0、5、10、20、50纳克/毫升)5孔及空白孔1孔,其余各孔加待检人血清样品,每孔加样100微升。置37℃,保持1小时,取出用洗涤液洗涤5次,拍干。除空白孔外,其余各孔加酶标记抗体100微升。置37℃,保持1小时,取出用洗涤液洗涤5次,拍子。各孔加底物显色液100微升。置37℃,保持15分钟,各孔加终止液50微升。
结果判断
(1)采用酶标检测仪,主波长492nm,副波长630nm,标准品0纳克/毫升校零,测定各孔吸光度值。以人重组多肽PSA2参考;
(2)以人重组多肽PSA2参考标准品标示值为横坐标,吸光度值为纵坐标,绘制标准曲线;
(3)以待检各孔的吸光度值查标准曲线得出相应的检测结果。
本发明的优点在于:以人多肽PSA2抗原得到相应的前列腺癌诊断试剂盒。该试剂盒操作简便,测试灵敏,重现性好。经大量试验检测获得前列腺癌的测定参考范围为(以每毫升人重组多肽PSA2纳克计):5ng/ml以下为阴性,以上为阳性。

实施例1、羊抗人重组多肽PSA2 IgG抗体制备
取健康雄性的50kg重成年绵羊,用人重组多肽PSA2纯化蛋白抗原/合成多肽进行免疫。第一次以抗原加卡介苗和福氏佐剂研匀,注射绵羊颈淋巴结和皮下肌肉组织,每二周一次,共免疫4-5次,抗原量5mg/次。免疫最后一次的二周后采血,羊抗人血清用饱和硫酸铵二级沉淀后上离子交换层析柱,得到羊抗人重组多肽PSA2IgG抗体,测定效价,冷冻备用。
实施例2、兔抗人人重组多肽PSA2 IgG抗体制备
取健康雄性的成年兔子(5kg),用人重组多肽PSA2纯化蛋白抗原/合成多肽进行免疫。第一次以抗原加卡介苗和福氏佐剂研匀,注射兔子皮下肌肉组织,每二周一次,共免疫4次,抗原量0.5mg/次。免疫最后一次的二周后采血。羊抗人血清用饱和硫酸铵二级沉淀后上离子交换层析柱,得到兔抗人人重组多肽PSA2IgG抗体,测定效价,冷冻备用。
实施例3、鼠抗人重组多肽PSA2 IgG抗体制备
取健康雄性的成年豚鼠,用人重组多肽PSA2纯化蛋白抗原/合成多肽进行免疫。第一次以抗原加卡介苗和福氏佐剂研匀,注射豚鼠皮下肌肉组织,每二周一次,共免疫4次,抗原量0.1mg/次。免疫最后一次的二周后采血。鼠抗人血清用饱和硫酸铵二级沉淀后上离子交换层析柱,得到鼠抗人重组多肽PSA2IgG抗体,测定效价,冷冻备用。
实施例4、抗体标记
取实施例1-3中任一方法获得的人重组多肽PSA2 IgG蛋白抗体采用常规过碘酸钠法标记辣根过氧化物酶,
(1)人重组多肽PSA2抗体(2mg/ml)0.9ml,加0.1mol pH9.5 NaAc;
(2)辣根过氧化物酶10mg溶解于1.0mil. 0mmolpH4.0NaAc。
(3)加0.1mol过碘酸钠0.1ml,1.6mmol乙二醇0.1ml于上述辣根过氧化物酶溶液。
(4)将上述辣根过氧化物酶溶液与抗体混合搅拌1-2小时。
(5)加0.1mol四氢硼化钠0.1ml于4~C静止30分钟。
(6)用50%饱和硫酸铵沉淀分离标记物。
(7)标记物用50%丙三醇------PBS溶解,并于冷冻储藏。
实施例5、包被抗体聚苯乙烯微孔板制备
取实施例4获得的标记的人重组多肽PSA2IgG抗体,工作浓度于50mM,pH9.5碳酸盐条件下加入聚苯乙烯微孔,每孔100微升,并于2-8℃条件放置约40小时。取出用生理盐水洗涤5次,烘干,用铝箔袋密封包装。
实施例6、人重组多肽PSA2试剂盒
试剂
包被抗体聚苯乙烯微孔板:固相抗人重组多肽PSA2抗体。
酶标记抗体:酶标记抗人重组多肽PSA2抗体、0.15molpH7.2磷酸盐一氯化钠溶液。
洗涤液(10X):8%氯化钠---0.5%Tween20。
显色缓冲液:0.05molpH5.0磷酸盐一柠檬酸溶液。
显色剂:邻苯二铵。
终止液:2mol硫酸溶液。
仪器

分析方法:双抗体夹心法原理定量测定
样品要求:人血清。
技术要求:(ELISA法)采用酶标检测仪,主波长492nm副波长630nm。
计算方法:以人多肽PSA2参考标准品标示值为横坐标,吸光度值为纵坐标准曲线。绘制标准曲线。以待检各孔的吸光度值查标准曲线得出相应的检测结果。
性能要求
物理性状:外观:各液体组份应清晰透明,包装无渗漏,密封袋无漏气。
试剂空白吸光度:在主波长492nm副波长630nm处,室温,吸光度应<0.15A。
分析灵敏度:15分钟,试剂空白;取人多肽PSA2标准每毫升0.16微克检测,检测结果±20%。
准确度:取人重组多肽PSA2标准每毫升0.33微克、检测结果±15%。
精密度:1.0微克 CV~15%。
测定范围:2.0微克范围内呈线性,线性误差应≤15%,测定吸光度值与标示值的相关系数r>0.98.
实验方法
物理性状:目测试剂盒各组份外观。
试剂空白吸光度:以含10%小牛血清生理盐水为空白样品进行检测,按规定操作,以显色底物空白校零,记录吸光度,重复三次取平均值。
分析灵敏度:检测人重组多肽PSA2标准每毫升5纳克,重复3次取平均值。
准确度:检测标准品,人重组多肽PSA2标准每毫升10纳克、,重复3次,取平均值。
精密度(重复性):取人多肽PSA2含量约每毫升20纳克的血清检测进行20次测定,(N=20),求出20次吸光度变化均值X和标准差S,按下式计算变异系数(CV),CV%=S/X
测定范围:取人重组多肽PSA2标准每毫升5纳克、10纳克、20纳克、50纳克、每一浓度重复检测3次,取平均值。
测试实例
基本参数:采用酶标检测仪,主波长492nm副波长630nm,以人重组多肽PSA2参考标准晶0微克/毫升校零,测定各孔吸光度值。
使用方法:
(1)每次实验每块包被抗体聚苯乙烯微孔板设人重组多肽PSA2参考标准晶(0、5纳克、10纳克、20纳克、50纳克/毫升)5孔及空白孔1孔,其余各孔加待检血清样品,每孔加样100微升。置37℃1小时,取出用洗涤液洗涤5次,拍干。
(2)除空白孔外,其余各孔加酶标记抗体100微升。置37℃,保持1小时,取出用洗涤液洗涤5次,拍干。
(3)各孔加底物显色液100微升。置室温避光,保持15分钟,加终止液50微升,吸光度(OD)0.80查表样品人重组多肽PSA2含量5.2纳克/ml。
结果判断:
(1)采用酶标检测仪,主波长492nm副波长630nm,以人重组多肽PSA2参考标准晶0纳克/毫升校零,测定各孔吸光度值。
(2)以人重组多肽PSA2参考标准晶标示值为横坐标,吸光度值为纵坐标,绘制标准曲线。
(3)样品OD(吸光度)0.80,查标准曲线图,样品人重组多肽PSA2值:5.2纳克/ml,此患者血清中人重组多肽PSA2含量5.2纳克/ml,前列腺癌阳性。

1.多肽PSA2在制备前列腺癌诊断试剂盒中的应用。
2.根据权利要求1所述的人多肽PSA2的氨基酸序列:

3.根据权利要求1所述的人多肽PSA2的氨基酸序列中任意短肽序列片断在制备前列腺癌诊断试剂盒中的应用。
4.根据权利要求3所述的人多肽PSA2的氨基酸序列,三种合成多肽PSA2的氨基酸序列分别为:
YYGRKKATLEWAMKNHLWGHC、MAHVPFGHYTVKTDHLVLLGC和MKNHLWGHALFLSSKMDPQTC。
5.一种多肽PSA2纯蛋白抗原。
6.一种多肽PSA2蛋白抗体,该抗体包括单克隆抗体和多克隆抗体。
7.如权利要求6所述的多肽PSA2蛋白抗体,该抗体为基础衍生出的人源化抗体在制备前列腺癌治疗药物中的应用。
8.一种前列腺癌特异性蛋白PSA2的检测试剂盒,包括包被有抗体的酶标板、酶标二抗、通用试剂Tween-20和显色底物四甲基联苯胺TMB,其特征在于该试剂盒的酶标板孔内为经包被液处理和阻滞液处理的PSA2融合蛋白抗体,另配备有辣根过氧化物酶HRP标记的兔抗PSA2多肽抗体。
9.多肽PSA2编码的cDNA及所在蛋白全长cDNA,对以其为基础设计合成的引物在制备前列腺癌诊断试剂盒中的应用。
10.对PSA2编码的cDNA及所在蛋白全长cDNA基础设计合成的反义核酸,siRNA对在前列腺癌诊断治疗中的应用。
