本申请系关于一种提升耐热性的葡萄糖氧化酶,其氨基酸序列系为将序列编号2第129个位置的谷氨酸突变成脯氨酸,及/或将第243个位置的谷酰氨酸突变成缬氨酸之氨基酸序列。
Description
【技术领域】
本申请系关于一种葡萄糖氧化酶,尤指一种提升耐热性的葡萄糖氧化酶。
【现有技术】
葡萄糖氧化酶(Glucose oxidase)属于一种氧化还原酶(EC 1.1.3.4)。在有氧的条件下,能够专一性地催化β-D-葡萄糖的氧化,进而生成葡萄糖酸(gluconic acid),同时产生过氧化氢(H2O2)。葡萄糖氧化酶首先是由Muller的实验室于1928年在黑曲霉(Aspergillus niger)的萃取物中发现它的存在。葡萄糖氧化酶广泛存在于动物、植物以及微生物的体内,当中以微生物来源的氧化酶之相关研究为最多,而微生物来源主要存在于黑曲霉以及青霉属(Penicillium spp.)菌种。目前大多数的工业用葡萄糖氧化酶也是利用上述两种天然菌株生产取得,然而这样的生产方式却存在着产量与活性偏低的问题。另外,它们也会同时生产许多除了氧化酶之外的杂蛋白,导致后续制程的纯化不易,最终造成生产成本的提高。因此,近年来也有研究利用其他的微生物系统生产氧化酶,尤其是在工业常用的真菌毕赤酵母(Pichia pastoris)系统中得到不错的进展。
葡萄糖氧化酶的应用范围十分广泛。在食品工业里,葡萄糖氧化酶可做为抗氧化的食品保鲜剂,用来保持蔬果等鲜度以及维持啤酒饮料的风味。葡萄糖氧化酶还可用在改良面团的筋度,提高面包的质量。医学领域中也可用葡萄糖氧化酶来检测血液中葡萄糖的含量。纺织工业里则利用葡萄糖氧化酶所产生的过氧化氢,其具有漂白(bleaching)功能,也可添加至衣物清洁剂。近年来,葡萄糖氧化酶更被应用在饲料工业中。由于葡萄糖氧化酶有减少氧气含量,降低环境中的pH值以及产生过氧化氢的作用,故可进而抑制某些细菌或真菌的生长。因此,添加葡萄糖氧化酶在饲料里面可改善动物的肠道环境。在其他方面,葡萄糖氧化酶甚至也有使用于生物能源(biofuel)当中的可能性。由此可知,葡萄糖氧化酶在不同工业的应用里都具有一定的重要性。因此,针对葡萄糖氧化酶的活性产量以及特性进行改良的研究也日益增多。
目前在许多相关的研究中,为了得到更佳的酶,除了在自然界中筛选出来之外,就是将现有的酶蛋白加以改造。本申请即欲藉由逻辑性地设计突变来提升葡萄糖氧化酶的耐热性,进而增加其在工业应用上的价值与潜力。
【发明内容】
本申请之目的在于改造现有葡萄糖氧化酶,利用结构分析及点突变技术,以有效提升葡萄糖氧化酶的耐热性,进而增加葡萄糖氧化酶的工业应用价值与潜力。
为达上述目的,本申请之一较广义实施方案为提供一种葡萄糖氧化酶,其氨基酸序列系为将序列编号2第129个位置的谷氨酸突变成脯氨酸,以及将第243个位置的谷酰氨酸突变成缬氨酸之氨基酸序列。
在一实施例中,编码该序列编号2之基因系从黑曲霉(Aspergillus niger)所分离出来的AnGOD基因。
在一实施例中,该葡萄糖氧化酶之氨基酸序列如序列编号10所示。
本申请之另一较广义实施方案为提供一种葡萄糖氧化酶,其氨基酸序列系为将序列编号2第129个位置的谷氨酸突变成脯氨酸之氨基酸序列。
在一实施例中,编码该序列编号2之基因系从黑曲霉(Aspergillus niger)所分离出来的AnGOD基因。
在一实施例中,该葡萄糖氧化酶之氨基酸序列如序列编号6所示。
本申请之又一较广义实施方案为提供一种葡萄糖氧化酶,其氨基酸序列系为将序列编号2第243个位置的谷酰氨酸突变成缬氨酸之氨基酸序列。
在一实施例中,编码该序列编号2之基因系从黑曲霉(Aspergillus niger)所分离出来的AnGOD基因。
在一实施例中,该葡萄糖氧化酶之氨基酸序列如序列编号8所示。

【实施方式】
体现本申请特征与优点的一些典型实施例将在后段的说明中详细叙述。应理解的是本申请能够在不同的态样上具有各种的变化,其皆不脱离本申请的范围,且其中的说明及图式在本质上系当作说明之用,而非用以限制本申请。
本申请的葡萄糖氧化酶(AnGOD)是从真菌黑曲霉(Aspergillus niger)菌株中所分离出来的基因。根据先前相关文献指出,葡萄糖氧化酶的最适活性约在37℃以及pH 6的条件之下。本申请将此葡萄糖氧化酶基因构筑于载体上,并且送入工业常用的毕赤酵母(Pichia pastoris)中表达其蛋白。为了提升此葡萄糖氧化酶的耐热性,本申请进一步分析其蛋白结构,挑选具改造潜力的氨基酸,再利用点突变技术(site-directed mutagenesis)进行改造。
蛋白质立体结构的稳定性与其耐热性有很大关联,而疏水性作用力又是影响蛋白稳定性的重要因素之一。因此,本申请进一步分析葡萄糖氧化酶的蛋白结构,试图藉由增加疏水性作用力以加强蛋白结构的稳定性,进而增进其耐热性。经分析后挑选出位在环状区(loop)上第129个氨基酸的谷氨酸(glutamate)以及位于二级结构β-平板(β-sheet)上第243个氨基酸的谷酰氨酸(glutamine)进行改造,利用点突变技术,分别单一突变成脯氨酸(proline)以及缬氨酸(valine),其后更将两个突变点组合成双突变点,而得到本申请具有提升耐热性的葡萄糖氧化酶。
以下将详述本申请改造葡萄糖氧化酶之方法及其所得到之改良葡萄糖氧化酶。
第1图显示野生型葡萄糖氧化酶AnGOD的核苷酸序列以及氨基酸序列。如第1图所示,AnGOD基因包含1749个碱基(核苷酸序列以序列编号1标示)以及583氨基酸(氨基酸序列以序列编号2标示)。首先,将AnGOD基因构筑在pPICZαA的载体上。接着将线性化之后的质粒DNA转殖到毕赤酵母中,再把转殖后的菌液涂于含有0.1mg/ml zeocin抗生素的YPD盘上,在30℃中培养2天,筛选出成功转化的转殖酵母细胞。之后挑选菌落,个别接种到YPD培养基中进行培养增生,再进一步地将菌体分离出来,转移到含有0.5%甲醇的BMMY培养基内,进而诱导目标蛋白的表达。经由离心将其菌液分离,收集含有被表达并且分泌至细胞外的目标蛋白之上清液。
AnGOD的三种突变基因则利用点突变技术取得,以野生型AnGOD基因做为模板进行聚合酶连锁反应,当中所用的突变引物列于第2图,其中E129P意指为AnGOD第129个位置之氨基酸由谷氨酸(glutamate)突变成脯氨酸(proline),且E129P突变引物序列以序列编号3标示,而Q243V为第243个位置之氨基酸由谷酰氨酸(glutamine)突变成缬氨酸(valine),且Q243V突变引物序列以序列编号4标示。因此,本申请利用点突变技术取得AnGOD的三种突变基因分别是E129P、Q243V以及E129P/Q243V。
第3图至第5图即显示本申请所构筑的三种突变型的核苷酸及氨基酸序列。第3图显示E129P突变型葡萄糖氧化酶的核苷酸序列以及氨基酸序列,其中核苷酸序列以序列编号5标示,氨基酸序列以序列编号6标示,且其第129个位置之氨基酸由谷氨酸(glutamate)突变成脯氨酸(proline)。第4图显示Q243V突变型葡萄糖氧化酶的核苷酸序列以及氨基酸序列,其中核苷酸序列以序列编号7标示,氨基酸序列以序列编号8标示,且其第243个位置之氨基酸由谷酰氨酸(glutamine)突变成缬氨酸(valine)。第5图显示E129P/Q243V突变型葡萄糖氧化酶的核苷酸序列以及氨基酸序列,其中核苷酸序列以序列编号9标示,氨基酸序列以序列编号10标示,且其第129个位置之氨基酸由谷氨酸(glutamate)突变成脯氨酸(proline),以及第243个位置之氨基酸由谷酰氨酸(glutamine)突变成缬氨酸(valine)。
接着,利用DpnI酶将原始的DNA模板移除掉。三种突变基因分别送入大肠杆菌内复制放大,再藉由DNA定序,确认突变序列的成功与否。最后,三种突变基因分别送入毕赤酵母中表达其突变蛋白,如同前述步骤。之后,再对野生型蛋白及突变蛋白分别进行葡萄糖氧化酶的活性测定及耐热性分析。
葡萄糖氧化酶的活性测定是藉由此酶在氧化葡萄糖的同时会产生过氧化氢,再经由过氧化氢酶(horseradish peroxidase)的催化下,邻联茴香胺(o-dianisidine)显色剂会与过氧化氢反应,进而被氧化后改变其呈色,藉此推算出葡萄糖氧化酶的活性。大致而言,2.5ml邻联茴香胺、0.3ml 18%葡萄糖以及0.1ml过氧化氢酶(90unit/ml)三者混合后,放入37℃水浴中预热。再加入0.1ml适当稀释过的酶液,反应3min后,再加入2ml硫酸以终止反应。最后在OD540nm波长下侦测其吸光值,进而计算出葡萄糖氧化酶的活性。
至于蛋白耐热性分析方面,将野生型蛋白及突变型蛋白的蛋白量定量之后,分别放在64℃、66℃、68℃以及70℃的环境中,进行热处理2min。随后放在冰上5min降温,再置于室温下5min回复。最后,同时测定没有进行热处理的样本之酶活性(作为对照组100%)以及热处理过的蛋白样本所残留的酶活性。
第6图显示野生型及突变型葡萄糖氧化酶的耐热性分析。如第6图所示,在不同的热处理温度(64℃-70℃)条件下,三种突变型蛋白E129P、Q243V以及E129P/Q243V的耐热性皆高于野生型蛋白。以68℃热处理的结果为例:野生型蛋白的残余活性剩下43.7%,而突变型蛋白E129P和Q243V的残留活性分别为49.8%以及50.2%,将两个突变点结合的双突变蛋白E129P/Q243V之残留活性则还有55.2%。由此可知,单一突变点E129P以及Q243V便能成功提升AnGOD的耐热性。而将两个突变点结合之后,更进一步地提高至少一成的耐热性。
综上所述,为了提升葡萄糖氧化酶AnGOD的耐热性,本申请进一步分析其蛋白结构,挑选具改造潜力的氨基酸,进行合理地改造。结果显示,所改造的三种突变型蛋白E129P、Q243V以及E129P/Q243V的耐热性皆高于野生型蛋白。因此,本申请成功地提升葡萄糖氧化酶AnGOD的耐热性,也进一步地增加此葡萄糖氧化酶的工业应用价值以及扩大其工业应用范围的可能性。

1.一种葡萄糖氧化酶,其氨基酸序列系为将序列编号2第129个位置的谷氨酸突变成脯氨酸,以及将第243个位置的谷酰氨酸突变成缬氨酸之氨基酸序列。
2.一种葡萄糖氧化酶,其氨基酸序列系为将序列编号2第129个位置的谷氨酸突变成脯氨酸之氨基酸序列。
3.一种葡萄糖氧化酶,其氨基酸序列系为将序列编号2第243个位置的谷酰氨酸突变成缬氨酸之氨基酸序列。
