Abstract
本发明公开了一种混合型热启动DNA聚合酶组合物、PCR扩增试剂盒及其应用,Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶,其中,抗Taq抗体能够与Taq DNA聚合酶特异性结合,突变型KOD DNA聚合酶与野生型KOD DNA聚合酶相比第147位的组氨酸突变为赖氨酸。实验结果表明,上述混合型热启动DNA聚合酶组合物,用于PCR扩增时错配率低,具有高效扩增与高保真性的双重特点,用于扩增时耐热好,无论是对较长的DNA片段还是低模板量的模板或是高GC含量的模板均具有较好扩增效果。

技术领域
本发明涉及分子生物技术领域,特别是涉及一种混合型热启动DNA聚合酶组合物、PCR扩增试剂盒及其应用。
背景技术
PCR扩增(聚合酶链式反应)是一种用于放大扩增特定的DNA片段的分子生物学技术,它可看作是生物体外的特殊DNA复制。其原理是利用DNA在体外摄氏95℃高温时变性会变成单链,低温(经常是60℃左右)时引物与单链按碱基互补配对的原则结合。再调温度至DNA聚合酶最适反应温度(72℃左右),DNA聚合酶沿着磷酸到五碳糖(5'-3')的方向合成互补链。
DNA聚合酶(DNA polymerase)是在复制DNA中起关键作用的酶。DNA聚合酶主要是以DNA为复制模板,将DNA由5’端开始复制到3’端的酶。目前DNA聚合酶大多数是纯化发酵产物得到,或对酶蛋白进行抗体或化学修饰以达到热启动。衡量DNA聚合酶保真性的一个通用标准是错配率,错配率越低保真性越好。用普通DNA聚合酶对样品进行扩增的错配率一般在10-5碱基/循环数。对于错配率要求非常严格的实验,如基因筛选、测序、突变检测等,普通DNA聚合酶远远不能满足要求。新出的KOD DNA聚合酶虽然能够在一定程度上降低错配率,然而错配率仍然有待进一步降低,且KOD DNA聚合酶的聚合能力一般较差,对较长片段的目标DNA很难扩增出来。此外,传统的DNA聚合酶对于模板量为1pg以下的扩增模板和GC含量较高扩增模板,扩增效果不好,不符合更高灵敏度和准确性的检测需求。
发明内容
基于此,有必要提供一种对较长片段的目标DNA、或低模板量以及高GC含量的扩增模板均具有较好扩增效果的DNA聚合酶。
此外,还有必要提供一种PCR扩增试剂盒及其应用。
一种混合型热启动DNA聚合酶组合物,包括Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶,其中,所述抗Taq抗体能够与所述Taq DNA聚合酶特异性结合,所述突变型KOD DNA聚合酶由野生型KOD DNA聚合酶的第147位的组氨酸突变为赖氨酸得到。
在一个实施方式中,所述突变型KOD DNA聚合酶包括:
(a)由SEQ ID No.1所示的氨基酸序列组成的蛋白质;或,
(b)在SEQ ID No.1所示的氨基酸序列中经过取代、缺失或添加一个或几个氨基酸且具有所述突变型KOD DNA聚合酶活性的由(a)衍生的蛋白质。
在一个实施方式中,所述Taq DNA聚合酶与所述突变型KOD DNA聚合酶的酶活比为1:10U~320U。
在一个实施方式中,所述Taq DNA聚合酶与所述突变型KOD DNA聚合酶的酶活比为1:80U~160U。
一种PCR扩增试剂盒,包括上述任一项所述的混合型热启动DNA聚合酶组合物。
在一个实施方式中,还包括GC扩增缓冲液,所述GC扩增缓冲液中含有40mmol/L~60mmol/L的Tris-HCl、50mmol/L~150mmol/L的KCl、2mmol/L~8mmol/L的MgCl2、20mmol/L~30mmol/L的(NH4)2SO4、体积分数为1%~3%的二甲基亚砜、体积分数为0.001%~0.01%的Tween20、体积分数为0.001%~0.01%的Triton-x-100和质量分数为0.5mg/mL~5mg/mL的BSA。
在一个实施方式中,还包括PCR缓冲液和PCR增强剂,所述PCR缓冲液中含有10mmol/L~30mmol/L的Tris-HCl、0.5mmol/L~5mmol/L的MgCl2、50mmol/L~80mmol/L的(NH4)2SO4以及40mmol/L~60mmol/L的KCl,所述PCR增强剂为甜菜碱。
上述任一项所述的PCR扩增试剂盒在扩增DNA中的应用,包括如下步骤:
将DNA扩增模板、PCR缓冲液以及混合型热启动DNA聚合酶组合物混合得到扩增体系;
将所述扩增体系置于PCR仪中进行PCR扩增。
在一个实施方式的应用中,所述DNA扩增模板中GC含量为65%以上。
在一个实施方式的应用中,所述扩增体系中的所述DNA扩增模板的含量为1pg以下。
实验结果表明,将Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶组合形成混合型热启动DNA聚合酶组合物,用于PCR扩增时错配率低,具有高效扩增与高保真性的双重特点,且出乎预料的是Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶用于扩增时可能发生了某种协同作用,使得混合型热启动DNA聚合酶组合物耐热好,从而使得配置扩增反应体系时,可在室温下配制,无需在冰上完成,操作简便。且无论是对较长的DNA片段还是低模板量的模板或是高GC含量的模板均具有较好扩增效果,能够用于基因筛选、测序、突变检测等。

具体实施方式
为使本发明的上述目的、特征和优点能够更加明显易懂,下面结合具体实施例及附图对本发明的具体实施方式做详细的说明。在下面的描述中阐述了很多具体细节以便于充分理解本发明。但是本发明能够以很多不同于在此描述的其它方式来实施,本领域技术人员可以在不违背本发明内涵的情况下做类似改进,因此本发明不受下面公开的具体实施的限制。
一实施方式的混合型热启动DNA聚合酶组合物,包括Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶,其中,抗Taq抗体能够与Taq DNA聚合酶特异性结合,突变型KOD DNA聚合酶由野生型KOD DNA聚合酶的第147位的组氨酸(H)突变为赖氨酸(K)得到。
需要说明的是,Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶可以预选混合,以混合组合物的形式存在。也可以是Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶可以分别储存,在使用的时候再按需要加入反应体系中。
在一个实施方式中,突变型KOD DNA聚合酶包括:(a)由SEQ ID No.1所示的氨基酸序列组成的蛋白质;或,(b)在SEQ ID No.1所示的氨基酸序列中经过取代、缺失或添加一个或几个氨基酸且具有该突变型KOD DNA聚合酶活性的由(a)衍生的蛋白质。该突变型KODDNA聚合酶对野生型的Taq DNA聚合酶进一步优化,将第407~766位的氨基酸以及第852~1388位氨基酸缺失,减少不必要的剪接体,并将第147位的组氨酸(H)突变为赖氨酸(K)。表达片段小,且仅突变一个位点,突变的位点少,容易制备。
可以理解,由于编码同一种氨基酸的密码子有多种,多肽的编码序列具有多态性及变异的特点。因此在SEQ ID No.1所示的氨基酸序列中经过取代、缺失或添加一个或几个氨基酸且具有本申请的突变型KOD DNA聚合酶活性的蛋白质,或没有明显的功能差异,也包括在本发明的范围内。
突变型KOD DNA聚合酶能够进步降低扩增的错配率。其错配率与野生型KOD DNA聚合酶相比能够降低了50%,在相对基数很低的条件下,显著提高了扩增的灵敏度。而且预料不到的发现,该突变型KOD DNA聚合酶与Taq DNA聚合酶、抗Taq抗体形成的混合型热启动DNA聚合酶组合物无论是对较长的DNA片段还是低模板量的模板或是高GC含量的模板均具有较好扩增效果,说明三者在用于扩增时发生了某种协同作用。
具体地,Taq DNA聚合酶是一种来源于嗜热菌Thermus aquaticus的高度热稳定的DNA聚合酶,其具有耐高温、较高的特异性等优良特性,被广泛应用在PCR扩增中。但是TaqDNA聚合酶在PCR第一个循环变性之前,有一个升温的过程(例如95℃),期间引物和模板会有一些非特异性配对,如果这时Taq酶发挥活性,就很容易产生非特异性扩增,由于循环初期模板量非常少,产生的非特异性条带经过后面的指数扩增,就会严重干扰目的片段的扩增,甚至导致特异性条带不能扩出。本实施方式的混合型热启动DNA聚合酶组合物中引入了抗Taq抗体,抗Taq抗体为Taq DNA聚合酶对应的抗体,能够与Taq DNA聚合酶特异性结合,在高温加热前,抗Taq抗体与Taq DNA聚合酶活性受到抑制,不会导致非特异性扩增。高温变性后,抗Taq抗体与Taq DNA聚合酶分离,Taq DNA聚合酶恢复催化活性,使特异性结合的引物延伸,从而大大提高了PCR反应的灵敏度及特异性。
在一个实施方式中,混合型热启动DNA聚合酶组合物使用时,Taq DNA聚合酶与抗Taq抗体的浓度为1~100:1~100,例如3:2。
在一个实施方式中,Taq DNA聚合酶包括:(a)由SEQ ID No.2所示的氨基酸序列组成的蛋白质;或,(b)在SEQ ID No.2所示的氨基酸序列中经过取代、缺失或添加一个或几个氨基酸且具有该Taq DNA聚合酶活性的由(a)衍生的蛋白质。具有上述氨基酸序列的TaqDNA聚合酶为高保真的Taq DNA聚合酶(HiFi TaqDNA polymerase),进一步降低扩增时的错配率。相应的,抗Taq抗体为抗高保真的Taq DNA聚合酶的抗体。
同样地,由于编码同一种氨基酸的密码子有多种,多肽的编码序列具有多态性及变异的特点。因此在SEQ ID No.2所示的氨基酸序列中经过取代、缺失或添加一个或几个氨基酸且具有本申请的Taq DNA聚合酶活性的蛋白质,或没有明显的功能差异,也包括在本发明的范围内。
在一个实施方式中,混合型热启动DNA聚合酶组合物使用时,Taq DNA聚合酶与突变型KOD DNA聚合酶的酶活比为1:10U~320U。该范围内的酶活比,扩增效果最好,获得的条带清晰,杂带少。
进一步地,使用时,Taq DNA聚合酶与突变型KOD DNA聚合酶的酶活比为1:80U~160U。该范围内的酶活比,扩增效果更好,获得的条带清晰,杂带更少。
上述混合型热启动DNA聚合酶组合物,包括Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶。该混合型热启动DNA聚合酶组合物至少具有如下有益效果:(1)混合型热启动DNA聚合酶组合物的耐热好,从而使得配置扩增反应体系时,可在室温下配制,无需在冰上完成,操作简便。(2)进步降低扩增的错配率,在相对基数很低的条件下,显著提高了扩增的灵敏度。(3)且突变型KOD DNA聚合酶仅需突变一个位点,突变的位点少,容易制备。(4)抗体结合的热启动方法克服了化学修饰和物理阻隔等方法造成的需要延长预变性时间和需要几个PCR循环才能充分释放DNA聚合酶活性的缺点,对酶的损伤最小,使用更为简便。(5)无论是对较长的DNA片段还是低模板量的模板或是高GC含量的模板均具有较好扩增效果。
此外,本申请还提供一实施方式的PCR扩增试剂盒,该PCR扩增试剂盒中含有上述一项的混合型热启动DNA聚合酶组合物。
混合型热启动DNA聚合酶组合物的特征可参见上文描述,在此不作赘述。
当然,PCR扩增试剂盒还可以包括包装盒、包装瓶等。
在试剂盒中,Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶可以预选混合,以混合酶的形式存在。也可以是Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶可以分别储存,在使用的时候再按需要加入反应体系中。
本实施方式中,Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶可以分别储存,在使用的时候再按需要加入反应体系中。
在一个实施方式中,PCR扩增试剂盒中还包括GC扩增缓冲液,该GC扩增缓冲液中含有40mmol/L~60mmol/L的Tris-HCl、50mmol/L~150mmol/L的KCl、2mmol/L~8mmol/L的MgCl2、20mmol/L~30mmol/L的(NH4)2SO4、体积分数为1%~3%的二甲基亚砜(DMSO)、体积分数为0.001%~0.01%的Tween20、体积分数为0.001%~0.01%的Triton-x-100和质量分数为0.5mg/mL~5mg/mL的BSA。
进一步地,该GC扩增缓冲液中含有50mmol/L的Tris-HCl(pH8.8)、100mmol/L的KCl、5mmol/L的MgCl2、25mmol/L的(NH4)2SO4、体积分数为2%的二甲基亚砜(DMSO)、体积分数为0.005%的Tween20、体积分数为0.002%的Triton-x-100和质量分数为1mg/mL的BSA。
上述配方的GC扩增缓冲液与混合型热启动DNA聚合酶组合物配合,能够扩增GC含量超过50%的扩增模板,甚至GC含量超过80%的扩增模板也能扩增。GC含量是指在DNA的4种碱基中,鸟嘌呤和胞嘧啶所占的比率。
在一个实施方式中,PCR扩增试剂盒中还包括PCR缓冲液。PCR缓冲液中含有10mmol/L~30mmol/L的Tris-HCl、0.5mmol/L~5mmol/L的MgCl2、50mmol/L~80mmol/L的(NH4)2SO4以及40mmol/L~60mmol/L的KCl。
进一步地,该PCR缓冲液中含有25mmol/L的Tris-HCl(pH8.0)、2mmol/L的MgCl2、60mmol/L的(NH4)2SO4以及50mmol/L的KCl。
上述配方的PCR缓冲液为反应提供合适的酸碱环境以及盐离子环境,与与混合型热启动DNA聚合酶组合物配合,实现高效率、高灵敏度的扩增。
在一个实施方式中,PCR扩增试剂盒中还包括PCR增强剂,PCR增强剂为甜菜碱。PCR增强剂能够增强PCR聚合能力以及特异性。
在一个实施方式中PCR扩增试剂盒中还包括dNTP等。
上述PCR扩增试剂盒中包括混合型热启动DNA聚合酶组合物,再配合精心配制的PCR缓冲液以及PCR增强剂等,能够用于基因筛选、测序、突变检测等。
上述任一项PCR扩增试剂盒在扩增DNA中的应用,包括如下步骤:
S110、将DNA扩增模板、PCR缓冲液以及混合型热启动DNA聚合酶组合物混合得到扩增体系。
S120、将扩增体系置于PCR仪中进行PCR扩增。
具体地,可参见普通PCR扩增的步骤,将混合型热启动DNA聚合酶组合物加入反应体系中,从而扩增DNA样本。
具体地,DNA扩增模板可来源于人基因组DNA、噬菌体DNA和丝状真菌里氏木霉DNA中的至少一种。Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶用于扩增时可能发生了某种协同作用,对于人基因组DNA、噬菌体DNA和丝状真菌里氏木霉DNA均能实现高灵敏度的扩增。
在一个实施方式的应用中,DNA扩增模板中的GC含量为50%以上。例如DNA扩增模板中的GC含量为50%~90%等。GC含量是指在DNA扩增模板中的4种碱基,鸟嘌呤和胞嘧啶所占的比率。
进一步地,DNA扩增模板中的GC含量为65%以上。例如DNA扩增模板中的GC含量为65%~85%等。
Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶用于扩增时可能发生了某种协同作用,对于高GC含量的DNA模板均能实现高灵敏度的扩增。
在一个实施方式的应用中,扩增体系中的DNA扩增模板的含量为1pg以下,例如10fg~1pg等。
Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶用于扩增时可能发生了某种协同作用,对于模板含量很低的DNA样本均能实现高灵敏度的扩增。
综上,将Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶组合形成混合型热启动DNA聚合酶组合物,用于PCR扩增时错配率低,具有高效扩增与高保真性的双重特点,且出乎预料的是Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶用于扩增时可能发生了某种协同作用,使得混合型热启动DNA聚合酶组合物耐热好,从而使得配置扩增反应体系时,可在室温下配制,无需在冰上完成,操作简便。且无论是对较长的DNA片段还是低模板量的模板或是高GC含量的模板均具有较好扩增效果,能够用于基因筛选、测序、突变检测等。
下面为具体实施例部分。
以下实施例中,如无特别说明,未注明具体条件的实验方法,通常按照常规条件,例如参见萨姆布鲁克、EF弗里奇、T曼尼阿蒂斯等(金冬雁,黎孟枫等译)所著的的分子克隆实验指南[M](北京:科学出版社,1992)中所述的条件或者试剂盒生产厂家推荐的方法实现。所有操作均采用本领域标准操作过程,所采用的试剂或载体等均为常规试剂或常规载体。
图中,Taq plus表示普通市售的Taq DNA聚合酶,Pfu-fusion表示普通市售的PfuDNA聚合酶,KD plus表示普通市售的KOD DNA聚合酶。KTaq MIX DNAPolymerase(KTq MIX)表示本申请中的混合型热启动DNA聚合酶组合物。
实施例1制备突变型KOD DNA聚合酶
根据SEQ ID No.1所示的氨基酸序列设计目的基因表达序列,并委托基因合成公司进行合成。本实施例委托苏州泓讯公司合成,并测序验证获得了特定位点突变的基因表达片段。将目的基因表达序列按照试剂盒推荐的方法转化到BL21(DE3)中进行诱导表达以及纯化,得到纯度90%以上的目的蛋白,蛋白大小约为90KD。纯化过程以及纯化后的各蛋白样品的SDS-PAGE图如图1所示,图上标记的“样品”表示诱导后收集的蛋白样品的电泳泳道,“流穿”表示流过纯化柱的样品,B1~B5、C1~C5以及D1表示纯化过程中用含不同浓度的咪唑的缓冲液冲洗收集的样品。A1、A5表示收集的纯化的样品。可以看出诱导BL21后,突变型KOD DNA聚合酶成功表达,透析后的样品的纯度高、杂质少。
制备Taq DNA聚合酶
根据SEQ ID No.2所示的氨基酸序列设计目的基因表达序列,并委托基因合成公司进行合成。本实施例委托苏州泓讯公司合成。将目的基因表达序列按照试剂盒推荐的方法转化到BL21(DE3)中进行诱导表达以及纯化,得到纯度90%以上的目的蛋白,蛋白大小约为100KD。诱导前后、纯化过程以及纯化后的各蛋白样品的SDS-PAGE图分别如图2~4所示,可以看出诱导BL21后,Taq DNA聚合酶成功表达,透析后的样品的纯度高、杂质少。
Taq DNA聚合酶对应的抗Taq抗体购自sigma公司。
突变型KOD DNA聚合酶、Taq DNA聚合酶、抗Taq抗体分别储存,备用。
实施例2PCR扩增试剂盒的应用
以25μL为例,按如下下表1所述PCR扩增体系进行溶液配置。

本发明推荐使用试剂盒中自带的2×PCR Enhancer(PCR增强剂),有利于扩增微量模板DNA。模板若GC含量超过50%建议使用GC buffer来进行扩增。
推荐PCR反应条件如下表2所示:

注意:选择合适的循环数以及合适的退火温度,避免得不到特异性扩增条带。
实施例3检测不同的PCR反应条件
以一个长度2.9Kb的DNA片段(PET-28a)为模板,设计以下扩增引物,
2.9Kb片段的扩增引物:
PET-28a-F:ATCCGGATATAGTTCCTCCT(如SEQ ID No.3所示)
PET-28a-R:CAGTATACACTCCGCTATGC(如SEQ ID No.4所示)
在统一其他一切反应条件的情况下,梯度变化PCR缓冲液中的MgC12的浓度(0mmol/L~5mmol/L),(NH4)2SO4的浓度(15mmol/L~65mmol/L),KCl的浓度(5mmol/L~55mmol/L),及其pH值(6.8~8.8),进行多组PCR反应,以PCR产物的琼脂糖电泳图来表征KTaq MIX DNA Polymerase(混合型热启动DNA聚合酶组合物)在PCR反应中的催化活性。其中,PCR产物条带越亮,就可以表示该组PCR反应的效率越高,即KTaq MIX DNA Polymerase的反应活性也越高,凝胶电泳图如图5。
根据各组PCR结果电泳图可知,混合型热启动DNA聚合酶组合物活性最高时,pH最佳值为8(25mmol/L的Tris-HCl),MgCl2最佳值为2mmol/L,(NH4)2SO4最佳值为60mmol/L,KCl最佳值为50mmol/L。
实施例4检测不同的酶活比
分别按照Taq DNA聚合酶与突变型KOD DNA聚合酶的按照酶活单位Taq:KODm=1U:10U/1U:20U/1U:40U/1U:80U/1U:160U/1U:320U的比例混合,换算成体积(μL)之比分别为1:6/1:12/1:24/1:48/1:96/1:192。取1μL按照实施例2推荐的PCR体系以及条件对Gp32进行扩增.
Gp32片段的引物:
Gp32-F:CGGGATCCATGTTTAAACGTAAAAGCA(如SEQ ID No.5所示)

凝胶电泳结果如图6显示,PCR产物条带越亮,就可以表示该组PCR反应的效率越高。可以看出在Taq DNA聚合酶与突变型KOD DNA聚合酶的酶活比(Taq:KODm)为1:10U~320U均能实现扩增,Taq DNA聚合酶与突变型KOD DNA聚合酶较好的的酶活比(Taq:KODm)为1:80U~160U。而单独的突变型KOD DNA聚合酶(KODm)对Gp32几乎没有扩增。综上KODm:Ptaq体积比为1:96(对应的Taq DNA聚合酶与突变型KOD DNA聚合酶的酶活比为1:160U)的扩增最好。
实施例5PCR扩增试剂盒
试剂盒中包括实施例1中Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶,以及PCR缓冲液、PCR增强剂、GC扩增缓冲液以及dNTP。
根据实施例2~4的的优化条件,配置的PCR缓冲液中含有25mmol/L的Tris-HCl(pH8.0)、2mmol/L的MgCl2、60mmol/L的(NH4)2SO4以及50mmol/L的KCl。PCR增强剂为甜菜碱。使用时按照Taq DNA聚合酶与突变型KOD DNA聚合酶的按照酶活单位Taq:KODm为1U:160U。
此外,使用时按照Taq DNA聚合酶与抗Taq抗体的浓度为3:2加入抗Taq抗体。
当扩增模板GC含量为50%以上加入GC缓冲液。GC扩增缓冲液含有50mmol/L的Tris-HCl(pH8.8)、100mmol/L的KCl、5mmol/L的MgCl2、25mmol/L的(NH4)2SO4、体积分数为2%的二甲基亚砜(DMSO)、体积分数为0.005%的Tween20、体积分数为0.002%的Triton-x-100和质量分数为1mg/mL的BSA。
dNTP内含有dATP、dGTP、dTTP、dCTP。
对实施例5的PCR扩增试剂盒进行性能评价
一、扩增速度的评价
以λDNA为模板分别扩增1kb~10kb片段时,退火时间设定为5sec.,延伸时间分别设定10sec和30sec,模板λDNA的加入量为1ng/50μL。PCR扩增条件:98℃10sec.55℃5sec.72℃10or 30sec(30cycle)。

取2μL的扩增产物进行1%的凝胶电泳,结果如下图7。结果显示延伸30sec的扩增效果好,条带清楚,说明混合型热启动DNA聚合酶组合物(KTaq MIX DNA Polymerase)扩增速率至少能够达到30sec/Kb,扩增效率高。对于8Kb、10Kb大片段的DNA模板依然能够很好的扩增。
二、热稳定性的评价
对里氏木霉HKMT基因按照实施例2推荐的PCR体系以及条件进行扩增,所用聚合酶分别有:
CompanyA/B/C商品化酶;
Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶按本申请实施例5推荐的配比形成的KTaq MIX DNA Polymerase(混合型热启动DNA聚合酶组合物);以及
Taq DNA聚合酶与市售的未突变的KOD DNA聚合酶形成的混合酶(相比混合型热启动DNA聚合酶组合物:KOD DNA聚合酶第147位的组氨酸未突变,未加入抗Taq抗体)。
然后将各组聚合酶放在37℃烘烤6天,在相同条件下对同一个基因里氏木霉HKMT基因按照上述PCR体系以及条件进行扩增。里氏木霉HKMT基因的引物:
HKMT-F:CTTGCACACGCATTGCAAGAGG(如SEQ ID No.19所示)
HKMT-R:GTTCTTATCACCATTCTTCCTCC(如SEQ ID No.20所示)
扩增产物的凝胶电泳图如下图8所示,1代表烘烤前,2代表烘烤后。相对于CompanyA/B/C商品化酶,烘烤前KTaq MIX DNA Polymerase(混合型热启动DNA聚合酶组合物)的扩增产物差不多。但是37℃烘烤6天后,混合型热启动DNA聚合酶组合物依然有较好的扩增效果,说明本申请的混合型热启动DNA聚合酶组合物具有良好的热稳定性,聚合酶可在室温下配制,无需在冰上完成,操作简便。
三、聚合能力的评价
以人基因组DNA为模板,分别用不同的引物扩增A/B/C/D不同基因片段。
A基因是NADPH基因

NADPH-R:CAAGTGCAAGCAGCCTTAGG(如SEQ ID No.22所示)

C基因是OBSCN基因

OBSCN-R:TACATGAATGATTATAGCA(如SEQ ID No.26所示)
D基因是MUC16基因
MUC16-F:AGGTAAGTGGGACTGGGCTG(如SEQ ID No.27所示)

A/B/C/D目的基因的大小分别为4.5Kb、5Kb、7.5Kb、9.0Kb。按照实施例2推荐的PCR体系以及条件进行扩增。模板DNA分别用三种不同浓度:50ng、5ng、0.5ng。取2μL的扩增产物进行1%的凝胶电泳,结果如下图9。结果显示,不同大小片段的基因片段均可被扩增出来,并且在模板浓度为0.5ng较低的条件也可以扩增出条带。
四、灵敏度的评价
以人基因组DNA为模板,扩增GPR98基因(大小6.0Kb)片段,
GPR98基因的引物:
GPR98-F:ATGGGTCCTATTGGAGCAGA(如SEQ ID No.29所示)
GPR98-R:AGGTCAGGAGATCGAGACCA(如SEQ ID No.30所示)
按照实施例2推荐的PCR体系以及条件进行扩增。模板DNA分别用七种不同浓度:10ng、1ng、100pg、10pg、1pg、100fg、10fg。
同时以其他三种商品化的高保真酶(Pfu-fusion、Taq plus、KD plus)同时扩增做对比。取2μL的扩增产物进行1%的凝胶电泳,结果如下图10。结果显示,KTaq MIXDNAPolymerase(KTq MIX)在模板浓度为100fg的条件也可以扩增出条带,而其他三种高保真酶却扩增不出条带。说明KTaq MIX DNA Polymerase具有超高的灵敏度。
五、对高GC含量模板的扩增效果的评价
以人基因组DNA为模板,模板DNA的GC含量分别为:47%、58%、60%、64%、67%、79%、84%。用不同的基因的引物来扩增片段,
扩增47%GC含量的引物:

按照实施例2推荐的PCR体系以及条件进行扩增,同时以其他三种商品化的高保真酶(Pfu-fusion、KD plus、Taq plus)同时扩增做对比。扩增时加入GC扩增缓冲液。取2μL的扩增产物进行1%的凝胶电泳,结果如下图11。
结果显示,KTaq MIX DNA Polymerase(KTq MIX)在模板GC含量为84%的条件也可以扩增出条带,而其他三种高保真酶却扩增不出条带。说明KTaq MIX DNA Polymerase(混合型热启动DNA聚合酶组合物)能够扩增困难模板。
六、保真性的评价
以λDNA为模板扩增大小为1Kb的基因片段。

按照实施例2推荐的PCR体系以及条件进行扩增,所用聚合酶分别有:

CompanyA高保真酶KOD DNA聚合酶(KOD);
CompanyA高保真酶Pfu DNA聚合酶(Pfu);
Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶按本申请实施例5推荐的配比形成的KTaq MIX DNA Polymerase(KTaq Mix,混合型热启动DNA聚合酶组合物)。
先切胶回收其目的片段,再用相对应的酶进行双酶切,同时将载体PUC19也进行双酶切,纯化回收其目的片段与载体。将具有同样粘性末端的片段连接,并且转化至JM109中,并且涂板于含有Xgal与IPTG的氨苄平板上,培养16h后,观察其平板生长情况并且计数,按照公式
f=-ln(F)/d×(bp)计算其保真性,
其中。F=White colonies/Total colonies(白色克隆/总克隆数),
d=number of template doublings(模板倍增数)。
统计结果图如下图12,结果显示:本申请的KTaq MIX DNA Polymerase(KTq MIX,混合型热启动DNA聚合酶组合物)的保真性可达到1.01×10-6,保真性比普通Taq酶高出50倍,比其他市面上高保真酶KOD DNA聚合酶以及Pfu DNA聚合酶高出2~4倍。

Claims (7)
1.一种混合型热启动DNA聚合酶组合物,其特征在于,由Taq DNA聚合酶、抗Taq抗体和突变型KOD DNA聚合酶组成,其中,所述抗Taq抗体能够与所述Taq DNA聚合酶特异性结合,所述突变型KOD DNA聚合酶由野生型KOD DNA聚合酶的第147位的组氨酸突变为赖氨酸得到;所述Taq DNA聚合酶与所述抗Taq抗体的浓度比为3:2;所述突变型KOD DNA聚合酶是由SEQ ID No.1所示的氨基酸序列组成的蛋白质;所述TaqDNA聚合酶与所述突变型KOD DNA聚合酶的酶活比为1U:(80U~160U)。
2.一种PCR扩增试剂盒,其特征在于,包括如权利要求1所述的混合型热启动DNA聚合酶组合物。
3.根据权利要求2所述的PCR扩增试剂盒,其特征在于,还包括GC扩增缓冲液,所述GC扩增缓冲液中含有40mmol/L~60mmol/L的Tris-HCl、50mmol/L~150mmol/L的KCl、2mmol/L~8mmol/L的MgCl2、20mmol/L~30mmol/L的(NH4)2SO4、体积分数为1%~3%的二甲基亚砜、体积分数为0.001%~0.01%的Tween20、体积分数为0.001%~0.01%的Triton-x-100和质量分数为0.5mg/mL~5mg/mL的BSA。
4.根据权利要求2所述的PCR扩增试剂盒,其特征在于,还包括PCR缓冲液和PCR增强剂,所述PCR缓冲液中含有10mmol/L~30mmol/L的Tris-HCl、0.5mmol/L~5mmol/L的MgCl2、50mmol/L~80mmol/L的(NH4)2SO4以及40mmol/L~60mmol/L的KCl,所述PCR增强剂为甜菜碱。
5.如权利要求2~4任一项所述的PCR扩增试剂盒在扩增DNA中的应用,其特征在于,包括如下步骤:
将DNA扩增模板、引物与PCR扩增试剂盒混合得到扩增体系;以及
将所述扩增体系置于PCR仪中进行PCR扩增。
6.根据权利要求5所述的应用,其特征在于,所述DNA扩增模板中GC含量为65%以上。
7.根据权利要求5所述的应用,其特征在于,所述扩增体系中的所述DNA扩增模板的含量为1pg以下。
