Abstract

技术领域
本发明涉及医药领域，具体涉及一种环孢素眼凝胶的处理工艺。
背景技术
环孢素A是一种从真菌中提取出来的含有11个氨基酸的环状多肽，是第三代高效免疫抑制剂，可用于治疗干眼病。
对于眼用制剂，国家要求按照注射剂管理，因此制剂的无菌保障十分关键。目前无菌保障主要有三个方面：除菌过滤工艺、无菌工艺和终端灭菌工艺。环孢素具有难溶性和热不稳定性，因此环孢素无菌制剂是一个难题。
目前主要是采用以下三种方法进行无菌处理：采购无菌原辅料，在无菌环境下操作；终产品湿热灭菌；终产品终端除菌工艺。第一种很难采购合格的无菌原辅料，且制剂工艺和车间环境全部采用无菌，生产成本很高，难度大；第二种终产品湿热灭菌，虽然可以保障制剂无菌合格，操作也比较好，但是环孢素本身不耐高温，导致环孢素产生较多的降解杂质，增加制剂安全风险，产生副作用；第三种终产品终端除菌工艺，仅可用于工艺小试工艺，很难商业化生产，因为乳剂凝胶剂粘度大，容易堵塞小孔径的滤膜，且过滤很慢，生产效率难以保证。
专利号为CN201410033737.3的“一种环孢素眼凝胶及其制备方法”，公开的用湿热灭菌的方式来保证无菌，重现检测发现，湿热灭菌导致环孢素产生降解杂质。
专利号为CN201510785005.4的“一种环孢素眼用乳剂组合物”的专利，是用无菌的原料和辅料配置制剂，该工艺在工业上应用十分困难。专利号为CN201610172271.4的“环孢素眼用乳剂”，该专利采用配置好的乳剂终端除菌过滤，对于粘稠的乳剂来说，很难通过0.22微米的无菌过滤器，因此不具备可操作性。综上，目前公开的各种无菌工艺，均不具备商业化生产的价值和可行性。
发明内容
本专利的目的之一是解决环孢素眼用高风险制剂的无菌问题。
有鉴于此，本发明提供一种环孢素A眼凝胶处理工艺，包含以下步骤：
步骤1：卡波姆加水后充分搅拌均质，制得的卡波姆基质溶胀后，再进行搅拌均质；
步骤2：基质湿热灭菌后冷却，进行抽真空脱气泡处理；用过滤后的氢氧化钠溶液调节基质pH为5.0-9.0；
步骤3：将蓖麻油聚烃氧酯(35)、环孢素A原料、1,2-丙二醇、水按照适宜比例充分混合在35～45℃水浴条件下制备成澄清溶液，过滤；
步骤4：滤液与步骤2所得基质混合均匀后，抽真空脱气泡；再一次过滤后，进行无菌灌装。
作为优选，步骤1所述溶胀时间为12-24小时；
作为优选，步骤2所述湿热灭菌为在115-121℃灭菌15-90min。更优选地，步骤2所述湿热灭菌为在117℃灭菌31min。
作为优选，步骤1所述卡波姆为卡波姆981。
作为优选，步骤2所述冷却为冷却至40℃以下；步骤2所述基质pH优选为7.0。
作为优选，步骤2所述过滤为经0.2um聚醚砜囊式过滤器过滤。
作为优选，步骤3所述澄清溶液在35-45℃制备。更优选地，所述澄清溶液在40℃水浴下制备。
作为优选，所述卡波姆与水的质量比为3:4500-5500。

与目前的技术相比，具有以下优点：
1、本发明所述工艺在加入环孢素加入卡波姆之前除菌过滤，相对于终产品终端除菌工艺，过滤介质粘度较小，便于工业化生产；
2、本发明所述工艺只需要局部无菌，相对于用无菌原辅料在全程无菌环境下，对原辅料的无菌要求低，便于采购和降低成本；
3、本发明所述工艺相对于终端灭菌，可以避免环孢素受热降解，减少产品杂质，提供安全性。
本发明所述工艺显著提高生产效率高，2万支只需要5个小时；生产的多批产品无菌合格，均一透明。
本发明所述工艺过滤效率高，吸附少，制剂粘度适宜，粘度对制剂的药效影响显著。对本发明所述工艺制备的制剂，对各项指标如性状、鉴别、酸碱度、渗透压、黏度、含量均匀度、含量、有关物质、无菌进行检测合格，提示具有良好的工业应用前景。

环孢素A眼凝胶工艺流程
1、卡波姆981基质的制备：
(1)向配液罐中加入适量的注射用水，加入处方量卡波姆981，搅拌、均质；
(2)向配液罐中加入适量的注射用水，搅拌、均质；
(3)基质静置溶胀，溶胀时间为12h；
(4)打开设备，搅拌、均质；
(5)115℃，基质湿热灭菌80min，同时搅拌和循环，基质冷却至40℃以下，抽真空脱气泡处理；
2、pH的调节：以氢氧化钠溶液，调节基质pH＝6.0，继续搅拌适当的时间；
3、环孢素A主药溶液和除菌过滤加入到配液罐：
(1)称取处方量的蓖麻油聚烃氧酯(35)置热水水浴中加热数分钟后缓缓加入处方量的环孢素A原料，边加边搅拌，在热水浴中至环孢素A原料完全溶解，加入处方量的1,2-丙二醇在热水浴中加热搅拌至澄清，加入适量的注射用水于35℃水浴中加热搅拌至澄清(其中蓖麻油聚烃氧酯(35)：环孢素A原料：1,2-丙二醇：注射用水＝15:1:24:34)，所需时间为30min，时间较长；
(2)用0.2umPES材料的囊式过滤器将(1)用蠕动泵加入至配液罐；
4、混合搅拌定重至批生产总量；
6、混合搅拌时间20分钟；
7、抽真空脱气泡30分钟；
8、中间体取样，检测pH、黏度、渗透压、有关物质及含量；
9、将中间体过30um聚丙烯囊式过滤器后，进行无菌灌装。
实施例2：
环孢素A眼凝胶工艺流程
1、卡波姆981基质的制备：
(1)向配液罐中加入适量的注射用水，加入处方量的卡波姆981，搅拌、均质；
(2)向配液罐中加入适量的注射用水，搅拌、均质；
(3)基质静置溶胀，溶胀时间为19h；
(4)打开设备，搅拌、均质；
(5)117℃，基质湿热灭菌31min，同时搅拌和循环，基质冷却至40℃以下，抽真空脱气泡处理；
2、pH的调节：以氢氧化钠溶液，调节基质pH＝7.0，继续搅拌数分钟；
3、环孢素A主药溶液和除菌过滤加入到配液罐：
(1)称取处方量的蓖麻油聚烃氧酯(35)置热水水浴中加热数分钟后缓缓加入处方量的环孢素A原料，边加边搅拌，在热水浴中至环孢素A原料完全溶解，加入处方量的1,2-丙二醇在热水浴中加热搅拌至澄清，加入适量的注射用水于40℃水浴中加热搅拌至澄清(其中蓖麻油聚烃氧酯(35)：环孢素A原料：1,2-丙二醇：注射用水＝20:1:44:64)，所需时间为10min，时间较实施例1少，节约时间；
(2)用0.2umPVDF材料的囊式过滤器将(1)用蠕动泵加入至配液罐；
4、混合搅拌定重至批生产总量；
6、混合搅拌时间20分钟；
7、抽真空脱气泡30分钟；
8、中间体取样，检测pH、黏度、渗透压、有关物质及含量；
9、将中间体过20um聚丙烯囊式过滤器后，进行无菌灌装。
实施例3：
环孢素A眼凝胶工艺流程
1、卡波姆981基质的制备：

(2)向配液罐中加入适量的注射用水，搅拌、均质；
(3)基质静置溶胀，溶胀时间为24h；
(4)打开设备，搅拌、均质；
(5)121℃，基质湿热灭菌20min，同时搅拌和循环，基质冷却至40℃温度以下，抽真空脱气泡处理；
2、pH的调节：以氢氧化钠溶液，调节基质pH＝9.0，继续搅拌数分钟；
3、环孢素A主药溶液和除菌过滤加入到配液罐：
(1)称取处方量的蓖麻油聚烃氧酯(35)置热水水浴中加热15min后缓缓加入处方量的环孢素A原料，边加边搅拌，在热水浴中至环孢素A原料完全溶解，加入处方量的1,2-丙二醇在热水浴中加热搅拌至澄清，加入适量的注射用水于45℃水浴中加热搅拌至澄清(其中蓖麻油聚烃氧酯(35)：环孢素A原料：1,2-丙二醇：注射用水＝25:1.5:46:66)，所需时间为8min，时间与实施例2相当；
(2)用0.2um聚丙烯材料的囊式过滤器将(1)用蠕动泵加入至配液罐；
4、混合搅拌定重至批生产总量；
6、混合搅拌时间20分钟；
7、抽真空脱气泡30分钟；
8、中间体取样，检测pH、黏度、渗透压、有关物质及含量；
9、将中间体过20um聚丙烯囊式过滤器后，进行无菌灌装。
实施例4：
环孢素A眼凝胶工艺流程
1、卡波姆981基质的制备：
(1)向配液罐中加入适量的注射用水，加入处方量的卡波姆981，搅拌、均质；
(2)向配液罐中加入适量的注射用水，搅拌、均质；
(3)基质静置溶胀，溶胀时间为24h；
(4)打开设备，搅拌、均质；
(5)121℃，基质湿热灭菌20min，同时搅拌和循环，基质冷却至40℃温度以下，抽真空脱气泡处理；
2、pH的调节：以氢氧化钠溶液，调节基质pH＝9.0，继续搅拌数分钟；
3、环孢素A主药溶液和除菌过滤加入到配液罐：
(1)称取处方量的蓖麻油聚烃氧酯(35)置热水水浴中加热后缓缓加入处方量的环孢素A原料，边加边搅拌，在热水浴中至环孢素A原料完全溶解，加入处方量的1,2-丙二醇在热水浴中加热搅拌至澄清，加入适量的注射用水于45℃水浴中加热搅拌至澄清(其中蓖麻油聚烃氧酯(35)：环孢素A原料：1,2-丙二醇：注射用水＝25:1.5:46:66)，所需时间为8min，时间与实施例2相当；
(2)用0.2um聚丙烯材料的囊式过滤器将(1)用蠕动泵加入至配液罐；
4、混合搅拌定重至批生产总量；
6、混合搅拌时间20分钟；
7、抽真空脱气泡30分钟；
8、中间体取样，检测pH、黏度、渗透压、有关物质及含量；
9、将中间体过10um聚丙烯囊式过滤器过滤，过滤器堵塞，生产终止。
实施例5：
本发明所述工艺制备的环孢素眼凝胶各项指标检测结果
从实施例1、2、3可知，实施例2是最优工艺。
以上所述仅是本发明的优选实施方式，应当指出，对于本技术领域的普通技术人员来说，在不脱离本发明原理的前提下，还可以做出若干改进和润饰，这些改进和润饰也应视为本发明的保护范围。
Claims (10)
1.一种环孢素A眼凝胶的处理工艺，其特征在于，包含以下步骤：
步骤1：卡波姆加水后充分搅拌均质，制得的卡波姆基质溶胀，再搅拌均质；
步骤2：基质湿热灭菌后冷却，进行抽真空脱气泡处理；用过滤后的氢氧化钠溶液调节基质pH为5.0-9.0；
步骤3：将蓖麻油聚烃氧酯(35)、环孢素A、1,2-丙二醇、水充分混合在35℃-45℃水浴条件下制备成澄清溶液；
步骤4：将澄清溶液过滤，滤液与步骤2所得基质混合均匀后，抽真空脱气泡；再次过滤后进行无菌灌装。
2.根据权利要求1所述的处理工艺，其特征在于，步骤1所述卡波姆与水的质量比为3:4500-5500；所述溶胀时间为12-24小时。
3.根据权利要求1所述的处理工艺，其特征在于，步骤2所述湿热灭菌为在115-121℃灭菌15-90min。
4.根据权利要求1所述的处理工艺，其特征在于，步骤2所述湿热灭菌为在117℃灭菌31min。
5.根据权利要求1所述的处理工艺，其特征在于，步骤2所述过滤为经0.2um聚醚砜囊式过滤器过滤。
6.根据权利要求1所述的处理工艺，其特征在于，步骤2所述基质pH为7.0。
7.根据权利要求1所述的处理工艺，其特征在于，步骤3所述澄清溶液在35-45℃制备，优选在40℃水浴制备。
8.根据权利要求1所述的处理工艺，其特征在于，步骤3所述蓖麻油聚烃氧酯(35)、环孢素A、1,2-丙二醇、水的质量比为20-30:1-2:44-48:64-68。
9.根据权利要求1所述的处理工艺，其特征在于，步骤3所述过滤为用0.2um PVDF材料的囊式过滤器过滤。
10.根据权利要求1所述的处理工艺，其特征在于，步骤4所述过滤为用20um聚丙烯囊式过滤器过滤。
