本发明涉及一种天然补铁软胶囊的制备方法,天然补铁剂包括乳铁蛋白、葡萄糖酸亚铁、维生素C、色拉油、白术粉以及炙甘草粉,按比例取各组分混合均匀,真空脱泡即得。软胶囊的制备步骤:首先取明胶加入水充分溶胀得到明胶液;其次将甘油与水混合,于水浴中加热至60‑80℃;最后加入明胶液,搅拌使其混合均匀,制成胶浆;将胶浆和权利要求3制得的天然补铁剂放入胶囊机制成胶囊;用乙醇洗去润滑油;烘干至水分合理,成丸。本发明中的天然补铁剂在葡萄糖酸亚铁中加入乳铁蛋白、白术粉、炙甘草粉等天然活性成分,能够提高缺铁性贫血的治疗效果,且胃肠道没有不良反应;同时制备的软胶囊产品整洁美观,容易吞服,色泽均匀,无明显气泡。

本发明涉及一种天然补铁软胶囊的制备方法,属于补铁药品及保健品技术领域。
背景技术
贫血是指全身循环血液中红细胞总量减少至正常值以下,但由于全身循环血液中红细胞总量的测定技术比较复杂,所以临床上一般指外周血中血红蛋白的浓度低于患者同年龄组、同性别和同地区的正常标准。贫血主要分为缺铁性贫血、出血性贫血、溶血性贫血、巨幼红细胞性贫血和再生障碍性贫血五大类,其中缺铁性贫血是临床上最为常见的一种类型。缺铁性贫血的特殊表现有:口角炎、舌乳突萎缩、舌炎,严重的缺铁可有匙状指甲(反甲),食欲减退、恶心及便秘。
血红蛋白由亚铁血红素和珠蛋白组成,亚铁血红素由铁和原卟啉合成,当缺铁或铁的吸收利用发生障碍时,亚铁血红素的合成减少,血红蛋白量降低,造成缺铁性贫血。此时通常的治疗为补充铁剂,对于一些患者来说,补铁剂是需要长期服用的。
目前常用的补铁剂为硫酸亚铁、乳酸亚铁( 如朴血口服液) 、葡萄糖酸亚铁( 如葡萄糖酸亚铁糖浆) 、琥珀酸亚铁( 如菲普利口服液) 、富马酸亚铁( 如小儿复方四维亚铁散)等。其中硫酸亚铁补铁剂吸收较差,胃肠道不良反应较大,口感较差;其余补铁剂吸收较好,胃肠刺激较小,但口感仍较差。
乳铁蛋白存在于牛乳清中,属于天然的转铁蛋白,乳铁蛋白通过它的氨基和羧基末端两个铁结合区域能高亲和的、可逆的与铁结合,并维持铁元素在一个较广的pH值范围内完成在十二指肠细胞的吸收和利用。除两个铁结合位点外,其他部位也可吸附铁离子。而且,如果体内铁处于过剩状态下,乳铁蛋白则可明显减缓铁对肠道的刺激作用。一是因为乳铁蛋白螯合了铁,避免了铁离子对肠道直接刺激作用.二是进食乳铁蛋白可以减少无机离子的摄入量。因此乳铁蛋白在人体内能够有效提高铁的吸收利用率,而且其代谢产物无刺激性,能够有效地提高人体免疫力,调节人体内代谢,消除贫血带来的免疫力低下,食欲不佳等不良影响。
白术、甘草为传统中药成分,具有健脾,燥湿利水,止汗,补中益气等功能;但目前并未发现将其用于补铁剂中,也没有关于其具有促进铁吸收作用的报道。
发明内容
针对现有技术的不足,本发明提供一种含天然活性成分的补铁制备方法,其通过在现有补铁剂基础上,通过添加乳铁蛋白等几种有效的天然活性成分,进一步提高了补血效果,大大改善了缺铁性贫血的治疗效果。

上述天然补铁剂,还包括以下重量份的成分:
白术粉 1-8
炙甘草粉 1-6。
所述色拉油可以选择为大豆色拉油。
上述天然补铁剂的制备方法,按比例取各组分混合均匀,真空脱泡即得。
将上述天然补铁剂制成软胶囊,其制备方法包括以下步骤:
溶胶:首先取明胶加入水充分溶胀得到明胶液;其次将甘油与水混合,于水浴中加热至60-80℃;最后加入明胶液,搅拌使其混合均匀,制成胶浆;
制丸:将胶浆和上述制得的天然补铁剂放入胶囊机制成胶囊;
洗涤:用乙醇洗去润滑油;
干燥:烘干至水分合理,成丸。
进一步的,所述溶胶步骤中明胶、甘油以及水的用量比为150-200mg :50-80mg:150-200ml,所述水的用量为明胶中加入的水和甘油中加入的水二者用量之和。
进一步的,所述溶胶步骤中搅拌时间为1-1.5小时。
进一步的,所述洗涤步骤中的乙醇为95%的乙醇。
进一步的,所述干燥步骤中烘干温度为30-35℃。
本发明中的天然补铁剂在葡萄糖酸亚铁中加入乳铁蛋白、白术粉、炙甘草粉等天然活性成分,能够提高缺铁性贫血的治疗效果,且胃肠道没有不良反应;同时制备的软胶囊产品整洁美观,容易吞服,色泽均匀,无明显气泡。
具体实施方式
以下结合具体实施例对本发明做进一步的说明,实施例1-8中均采用以下工艺制备:
配料:按比例取各组分混合均匀,真空脱泡得到内容物;
溶胶:取明胶150-200mg加入适量水充分溶胀,将部分水与甘油50-80ml混合,于水浴中加热至60-80℃,加入明胶液,搅拌使其混合均匀约1-1.5h,制成胶浆;水的总用量是150-200ml;
制丸:将胶浆和内容物放入胶囊机制成胶囊;
洗涤:用95%乙醇洗去润滑油;
干燥:30-35℃烘干至水分合理,成丸。
实施例1
将乳铁蛋白80kg、葡萄糖酸亚铁8kg、维生素C 1kg、色拉油50kg搅拌均匀,最后灌装制成软胶囊,胶囊每粒500mg。
实施例2
将乳铁蛋白120kg、葡萄糖酸亚铁20kg、维生素C 5kg、色拉油80kg搅拌均匀,最后灌装制成软胶囊,胶囊每粒500mg。
实施例3
将乳铁蛋白80kg、葡萄糖酸亚铁8kg、白术1kg、炙甘草粉1kg、维生素C 1kg、色拉油50kg搅拌均匀,最后灌装制成软胶囊,胶囊每粒500mg。
实施例4
将乳铁蛋白120kg、葡萄糖酸亚铁20kg、白术8kg、炙甘草粉6kg、维生素C 5kg、色拉油80kg搅拌均匀,最后灌装制成软胶囊,胶囊每粒500mg。
实施例5
将乳铁蛋白100kg、葡萄糖酸亚铁14kg、白术4kg、炙甘草粉3kg、维生素C 0.5kg、色拉油65kg搅拌均匀,最后灌装制成软胶囊,胶囊每粒500mg。
实施例6
将乳铁蛋白90kg、葡萄糖酸亚铁5kg、白术4kg、炙甘草粉1kg、维生素C 1kg、色拉油45kg搅拌均匀,最后灌装制成软胶囊,胶囊每粒500mg。
实施例7
将乳铁蛋白90kg、葡萄糖酸亚铁5kg、白术3kg、炙甘草粉2kg、维生素C 1kg、色拉油45kg搅拌均匀,最后灌装制成软胶囊,胶囊每粒500mg。
实施例8
将乳铁蛋白85kg、葡萄糖酸亚铁4kg、白术粉3kg、炙甘草粉1.5kg、维生素C 1.5kg、色拉油50kg搅拌均匀,最后灌装制成胶囊,胶囊每粒500mg。
毒性试验
本实验采用健康 SD 大鼠 ,口服给药实施例 1 -8制备的药物剂量梯度均为 0.01g/kg 以及0.1g/kg,连续给药30天,经观察大鼠给药期间及停药后1周之内动物生长状态、活动饮食、血液学、血液生化学、脏器组织结构以及尿常规等均无明显影响,表明临床用药安全可靠。
效果试验
分别取实施例1-8的胶囊作为实验品,互为对照。
缺铁性贫血病人200例,年龄25-65岁,平均年龄为35岁。随机分组,实施例1-8各25例,贫血判断标准男性血红蛋白<120g/l,女性血红蛋白<110g/l。
疗效评价
治愈 :贫血、口角炎、气虚乏力所有症状消失,并经医生确诊已经痊愈 ;
有效 :贫血、口角炎症状减轻、气虚乏力部分症状消失或好转 ;
无效 :贫血、口角炎、气虚乏力所有症状没有任何改善,甚至更加严重 ;
复发 :治愈者在停药后 6 个月内重新患上原有病症。
试验数据统计分析
试验全部结束后,将试验数据统计分析如下表所示 :
由上表可见 ,实施例1-8制备的补铁剂软胶囊均具有较好的补血效果,并且实施例3-8中加入了中药成分白术和炙甘草后,相比于实施例1和2治愈率和总有效率均明显提高;实施例5为最优的实施例,其补血效果最佳。
Claims (9)
1.一种天然补铁剂,其特征在于,包括以下重量份的成分:
乳铁蛋白 80-120
葡萄糖酸亚铁 8-20
维生素C 0.5-5
色拉油 50-80。
2.根据权利要求1所述的天然补铁剂,其特征在于,还包括以下重量份的成分:
白术粉 1-8
炙甘草粉 1-6。
3.根据权利要求1或2所述天然补铁剂,其特征在于,所述色拉油为大豆色拉油。
4.权利要求-3任一所述天然补铁剂的制备方法,其特征在于,按比例取各组分混合均匀,真空脱泡即得。
5.一种天然补铁软胶囊的制备方法,其特征在于,包括以下步骤:
溶胶:首先取明胶加入水充分溶胀得到明胶液;其次将甘油与水混合,于水浴中加热至60-80℃;最后加入明胶液,搅拌使其混合均匀,制成胶浆;
制丸:将胶浆和权利要求3制得的天然补铁剂放入胶囊机制成胶囊;
洗涤:用乙醇洗去润滑油;
干燥:烘干至水分合理,成丸。
6.根据权利要求5所述的天然补铁软胶囊的制备方法,其特征在于,所述溶胶步骤中明胶、甘油以及水的用量比为150-200mg :50-80mg:150-200ml,所述水的用量为明胶中加入的水和甘油中加入的水二者用量之和。
7.根据权利要求5所述的天然补铁软胶囊的制备方法,其特征在于,所述溶胶步骤中搅拌时间为1-1.5小时。
8.根据权利要求5所述的天然补铁软胶囊的制备方法,其特征在于,所述洗涤步骤中的乙醇为95%的乙醇。
9.根据权利要求5所述的天然补铁软胶囊的制备方法,其特征在于,所述干燥步骤中烘干温度为30-35℃。
