Abstract
本发明公开了一种治疗灰指甲的乳剂及其制备方法,该乳剂中含有以下物质:桉叶油、莪术油、鸦胆子油、丁香油酚、自乳化复合型乳化剂AC-402、橄榄乳化剂、尿素、水杨酸、樟脑、冰片、CMC-Na、海藻酸钠、氮酮、硼酸、甘油、羟丙基甲基纤维素钠、1,2-丙二醇、茴香、苦参、5%尼泊金乙酯丙二醇溶液。本发明所制备的治疗灰指甲乳液功能性强,毒性、刺激性小。鸦胆子油可抑制真菌繁殖,有效的阻止真菌感染。苦参提取液则具有消炎、抗菌功效。配方中的各成分相辅相成。相比传统药物,治疗效果更好,渗透性更强,复发性更低,从根本上治疗由各种真菌所引起的灰指甲。
Description
技术领域
本发明涉及一种治疗灰指甲乳剂及其制备方法,属于药剂学领域。
背景技术
现实生活中因真菌感染所致的灰指甲成为越来越多人的烦恼,而长期以来,灰指甲的诊断治疗方式都是比较单一的,治疗效果都不甚理想,反复复发成为治疗难题。
灰指甲发病的原因主要有三大类:个人因素、环境因素、疾病因素。
一、个人因素
1、不注意个人卫生;2、患有手足癣;3、长期与水或化学物直接接触;4、营养不良者;5、指甲受外伤或冲击,可能导致真菌侵入;6、经常与灰指甲(甲癣)患者接触。
二、环境因素
气温高、湿度大是癣病多发的重要条件。因此,夏季、雨季的癣病比冬季、旱季多且重。患者的工作、生活环境也不可忽视。在高温、高湿车间、特别是必须穿着厚重工作服才能生产而通风又差的环境易患癣病,若无洗澡设备或未能及时沐浴,则癣病发病率将明显提高。
三、疾病因素
某些慢性病如营养不良、甲状腺功能亢进症、糖尿病患者的癣病很多见,症状也偏重。一些重症,如艾滋病、晚期癌症患者,不仅真菌性癣病发病率高,而且癣病分布广泛,表现奇特,十分顽固难愈。
将毒性作用更小,抑制真菌能力更强的中药成分添加到制备好的渗透性更强的乳剂中将能有效的解决灰指甲所带来的烦恼。
发明内容
本发明的目的在于提供一种毒性作用小、抑制真菌能力强且渗透性强的治疗灰指甲的乳剂及其制备方法。
本发明的一种治疗灰指甲的乳剂,其中包含有以下用量的物质:
上述所述的治疗灰指甲的乳剂中,还包含有下述用量的物质:
上述所述的治疗灰指甲的乳剂中,所述的苦参提取液通过以下方法制备得到:取苦参2-15g,粉碎成粗粉,分三次提取:第一次加150-200ml0.1%-0.5%的盐酸浸泡5-30min,用回流提取法提取0.5-2h,收集提取液,第二次加入130-150ml0.1%-0.5%的盐酸提取0.5-2h,收集提取液,第三次加入100-120ml0.1%-0.5%的盐酸提取0.5-1h,每次滤过,合并三次滤液,混合浓缩至10-90ml。其中,0.1%-0.5%的盐酸为体积浓度。
上述所述的治疗灰指甲的乳剂中,所述的茴香提取液通过以下方法制备得到:取茴香2-15g,粉碎成粗粉,分三次提取:第一次加入200-300ml70%-80%乙醇浸泡5-30min,用回流提取法提取0.5-1.5h,收集提取液,第二次加入150-200ml70%-80%乙醇提取0.2-1h,收集提取液,第三次加入100-150ml70%-80%乙醇提取0.2-1h,每次滤过,合并三次滤液,混合浓缩至10-90ml。其中,70%-80%乙醇为体积浓度。

本发明所述的治疗灰指甲的乳剂的制备方法,包括下述步骤:
a)称取海藻酸钠,将其分散于10-15ml蒸馏水中,充分溶胀成透明的凝胶A;
b)称取羧甲基纤维素钠,将其分散于6-10ml蒸馏水中,充分溶胀成透明的凝胶B;
c)称取羟丙基甲基纤维素,将其分散于6-10ml蒸馏水中,充分溶胀成透明的凝胶C;
d)称取桉叶油、莪术油、鸦胆子油、丁香油酚、自乳化复合型乳化剂AC-402、橄榄乳化剂于烧杯1中,60-90℃水浴加热,作为油相,备用;
e)量取蒸馏水1-10ml于烧杯2中,60-90℃水浴加热,作为水相,备用;
f)将烧杯2中的蒸馏水呈细流状加入烧杯1中,边加边朝同一个方向搅拌,形成乳剂基质a,备用;
g)往乳剂基质a中加入凝胶A、凝胶B、凝胶C,形成基质b,备用;
h)将尿素、水杨酸、硼酸分散于1-6ml蒸馏水中,形成溶液a,备用;
i)将樟脑、冰片分散于1-5ml75%乙醇中,形成溶液b,备用;
j)往基质b中加入溶液a、溶液b、氮酮、丙三醇、1,2-丙二醇、茴香提取液、苦参提取液、5%尼泊金乙酯醇溶液,搅拌混合均匀,即得治疗灰指甲的乳剂。
本发明以自乳化复合型乳化剂AC-402和橄榄乳化剂为乳化剂,以羧甲基纤维素钠、海藻酸钠、羟丙基甲基纤维素为乳化稳定剂,以丙三醇、1,2-丙二醇为助乳化剂。所述5%尼泊金乙酯醇溶液是为重量浓度,其中醇可以为乙醇、丙二醇、甘油等。
本发明中的各种成分如下:
桉叶油有对抗病毒的作用,对呼吸道疾病最有效,能缓和发炎现象,使鼻粘膜舒适。特别是对鼻窦炎、流行性感冒、咳嗽、鼻粘膜炎、气喘与肺结核特别有助益。对烫伤有明显功效,预防细菌、真菌滋生及随之而来的蓄脓,促进新组织的建构。割伤、伤口、溃疡与发炎状况,使用尤加利后均反应良好。
鸦胆子油可渗透深处皮肤,抑制真菌。其主要成分油酸,它能够抑制拓扑异构酶(TOPOII)活性,从而抑制细胞DNA的合成及生长,阻断真菌的增殖。
莪术油抗菌谱广,对皮肤丝状菌、念珠菌、酵母菌、黑色丝状菌、曲霉属、青霉属、放线菌等有抗菌作用。同时对某些革兰阳性菌也具有抗菌活性。其作用机制为作用于致病菌的细胞膜,改变其通透性,阻止营养摄取,导致其死亡。
丁香油酚对食品常见污染菌金黄色葡萄球菌、大肠杆菌、枯草芽胞杆菌、汉逊酵母、黑曲霉、青霉等有很强的抑制作用。丁香油酚少量滴人龋齿腔,除发挥消毒作用外,并破坏其神经,从而减轻牙痛。对表皮真菌也能抑制。
尿素是一种很好用的保湿成分,它就存在于肌肤的角质层当中,属于肌肤天然保湿因子NMF的主要成分。对肌肤来说,尿素具有保湿以及柔软角质的功效,从而辅助有效成分的渗入。
水杨酸可刺激皮肤、黏膜,因能与机体组织中的蛋白质发生反应能使角膜增殖后剥离。
苦参中富含苦参碱,苦参碱具有抗病原体作用,对结核杆菌也有抑制作用。
茴香中含挥发油、有机酸类、黄酮类、茴香醚、茴香醛等有效成分,对大肠杆菌、痢疾杆菌等都有很好的抑制作用。可帮助杀灭胃肠道和表皮的有害病菌,促进炎症及溃疡的痊愈。
樟脑涂于皮肤有温和的刺激及防腐作用。用力涂擦有发赤作用;轻涂则类似薄荷。樟脑还对常见致病真菌有抑制作用,可改善局部血液循环,能促进粘膜分泌。
冰片可抑制或杀灭金黄色葡萄球菌、乙型溶血性链球菌、绿色链球菌等临床常见细菌,冰片对红色癣菌等5种常见真菌亦有抗菌作用,其作用原理与冰片易透过细胞壁,引起细胞膜损伤,破坏及改变其通透性有关。
本发明所制备的治疗灰指甲的乳剂功能性强,毒性、刺激性小。鸦胆子油可抑制真菌繁殖,有效的阻止真菌感染。苦参提取液则具有消炎、抗菌功效。配方中的各成分相辅相成。相比传统药物,治疗效果更好,渗透性更强,复发性更低,从根本上治疗由各种真菌所引起的灰指甲。
具体实施方式
下述实施例是对于本发明内容的进一步说明以作为对本发明技术内容的阐释,但本发明的实质内容并不仅限于下述实施例所述,本领域的普通技术人员可以且应当知晓任何基于本发明实质精神的简单变化或替换均应属于本发明所要求的保护范围。
1茴香提取液的制备
取苦参15g,粉碎成粗粉,分三次提取:第一次加入200ml0.2%盐酸浸泡30min,用回流提取法提取2h,过滤收集提取液,第二次向滤渣中加入150ml0.2%盐酸提取2h,过滤收集提取液,第三次向滤渣中加入120ml0.2%盐酸提取1h,合并三次滤液,混合浓缩至90ml。
2苦参提取液的制备
取茴香15g,粉碎成粗粉,分三次提取:第一次加入300ml75%乙醇浸泡30min,用回流提取法提取1.5h,收集提取液,第二次向滤渣中加入200ml75%乙醇提取1h,收集提取液,第三次向滤渣中加入150ml75%乙醇提取1h,每次滤过,合并三次滤液,混合浓缩至90ml。
实施例1
治疗灰指甲乳剂的制备方法
a)称取海藻酸钠0.04g,将其分散于10ml蒸馏水中,充分溶胀成透明的凝胶A:
b)称取羧甲基纤维素钠0.05g,将其分散于6ml蒸馏水中,充分溶胀成透明的凝胶B:
c)称取羟丙基甲基纤维素0.06g,将其分散于6ml蒸馏水中,充分溶胀成透明的凝胶C:
d)称取桉叶油1.0g、莪术油0.7g、鸦胆子油0.6g、1.0g自乳化复合型乳化剂AC-402、0.5g橄榄乳化剂于烧杯1中,90℃水浴加热,作为油相,备用;
e)量取蒸馏水8ml于烧杯2中,90℃水浴加热,作为水相,备用;
f)等油相、水相达到相同温度,将烧杯2中的蒸馏水呈细流状加入烧杯1中,边加边朝同一个方向搅拌,形成乳剂基质a,备用;
g)往乳剂基质a中加入凝胶A、凝胶B、凝胶C,形成基质b,备用;
h)将0.5g尿素、0.3g水杨酸、0.2g硼酸分散于6ml蒸馏水中,形成溶液a,备用;
i)将0.5g樟脑、0.5g冰片分散于5ml75%乙醇中,形成溶液b,备用;
j)往基质b中加入溶液a、溶液b、氮酮0.1g、丙三醇5ml、1,2-丙二醇5ml、茴香提取液5ml、苦参提取液5ml、5%尼泊金乙酯丙二醇溶液4ml,搅拌混合均匀,即得治疗灰指甲的乳剂。
实施例2
治疗灰指甲乳剂的制备方法
a)称取海藻酸钠0.05g,将其分散于12ml蒸馏水中,充分溶胀成透明的凝胶A。
b)称取羧甲基纤维素钠0.10g,将其分散于10ml蒸馏水中,充分溶胀成透明的凝胶B;
c)称取羟丙基甲基纤维素0.12g,将其分散于10ml蒸馏水中,充分溶胀成透明的凝胶C;
d)称取桉叶油0.8g、莪术油0.6g、鸦胆子油0.6g、1.0g自乳化复合型乳化剂AC-402、0.4g橄榄乳化剂于烧杯1中,80℃水浴加热,作为油相,备用;
e)量取蒸馏水10ml于烧杯2中,80℃水浴加热,作为水相,备用;
f)等油相、水相达到相同温度,将烧杯2中的蒸馏水呈细流状加入烧杯1中,边加边朝同一个方向搅拌,形成乳剂基质a,备用;
g)往乳剂基质a中加入凝胶A、凝胶B、凝胶C,形成基质b,备用;
h)将0.4g尿素、0.2g水杨酸、0.1g硼酸分散于5ml蒸馏水中,形成溶液a,备用;
i)将1g樟脑、0.5g冰片分散于5ml75%乙醇中,形成溶液b,备用;
j)往基质b中加入溶液a、溶液b、氮酮0.05g、丙三醇4ml、1,2-丙二醇4ml、茴香提取液4ml、苦参提取液4ml、5%尼泊金乙酯丙二醇溶液3ml,搅拌混合均匀,即得治疗灰指甲的乳剂。
实施例3
治疗灰指甲乳剂的制备方法
a)称取海藻酸钠0.06g,将其分散于15ml蒸馏水中,充分溶胀成透明的凝胶A;
b)称取羧甲基纤维素钠0.07g,将其分散于10ml蒸馏水中,充分溶胀成透明的凝胶B;
c)称取羟丙基甲基纤维素0.12g,将其分散于10ml蒸馏水中,充分溶胀成透明的凝胶C;
d)称取桉叶油1.0g、莪术油0.7g、鸦胆子油0.6g、0.8g自乳化复合型乳化剂AC-402、0.5g橄榄乳化剂于烧杯1中,90℃水浴加热,作为油相,备用;
e)量取蒸馏水8ml于烧杯2中,90℃水浴加热,作为水相,备用;
f)等油相、水相达到相同温度,将烧杯2中的蒸馏水呈细流状加入烧杯1中,边加边朝同一个方向搅拌,形成乳剂基质a,备用;
g)往乳剂基质a中加入凝胶A、凝胶B、凝胶C,形成基质b,备用;
h)将0.4g尿素、0.2g水杨酸、0.2g硼酸分散于6ml蒸馏水中,形成溶液a,备用;
i)将0.5g樟脑、0.5g冰片分散于5ml75%乙醇中,形成溶液b,备用;
j)往基质b中加入溶液a、溶液b、氮酮0.1g、丙三醇5ml、1,2-丙二醇3ml、茴香提取液3ml、苦参提取液3ml、5%尼泊金乙酯丙二醇溶液4ml,搅拌混合均匀,即得治疗灰指甲的乳剂。
试验例1
实施例1-3所得乳剂的理化指标如下:
1性状
本品为淡棕黄色,粘稠度较稀薄、细腻均匀,涂展性良好的乳液。
2pH值检查
取本品,用pH计测定,测得pH值为6.8-7.0之间。
3冷热试验
将乳剂封装于透明乳液瓶中,于4℃的冰箱冷藏一个月观察无分层现象。于55℃的恒温箱内放24h,乳液无分层、变味等现象。
4离心试验
将乳剂封装于具塞试管中3000r/min离心20min,无分层现象。
5室温放置实验
将实施例乳剂置于乳液瓶中,室温静置考察6个月,无分层现象,用后感觉无变化,气味无变化。
6刺激性试验、过敏性实验
小白鼠背部剪去毛,分别涂抹按实施例制备的乳剂于剪去毛的部位,和没涂部位进行比较,结果没有刺激性和过敏反应。
取实施例适量涂于志愿者(16-50岁,80人)手面上,过30分钟后观察,均没出现发红,发疹,起泡现象。
试验例2
本发明实施例1-3所得治疗灰指甲乳剂的综合效果考察。
以下利用疗效来评价实施例1-3制造出的治疗灰指甲乳剂的功效。采用民意调查评分法,选取120名20-60岁年龄段的患者作为试用对象。任意等分三组,分别用实施例1、2、3制造出的治疗灰指甲乳剂涂抹于患处。每天使用两次,使用时间为三个月。各项使用效果总分为5分:5分为最高分,表示很好,非常满意;4分为较好;3分为可以接受;3分以下为不好,不能接受。以下为各项平均得分。结果见表1:

综上所述,本品气味佳,细腻、均匀,治疗灰指甲效果较好,且刺激性小。

5.权利要求2-4任一项所述的治疗灰指甲的乳剂的制备方法,包括下述步骤:
a)称取海藻酸钠,将其分散于10-15ml蒸馏水中,充分溶胀成透明的凝胶A;
b)称取羧甲基纤维素钠,将其分散于6-10ml蒸馏水中,充分溶胀成透明的凝胶B;
c)称取羟丙基甲基纤维素,将其分散于6-10ml蒸馏水中,充分溶胀成透明的凝胶C;
d)称取桉叶油、莪术油、鸦胆子油、丁香油酚、自乳化复合型乳化剂AC-402、橄榄乳化剂于烧杯1中,60-90℃水浴加热,作为油相,备用;
e)量取蒸馏水1-10ml于烧杯2中,60-90℃水浴加热,作为水相,备用;
f)将烧杯2中的蒸馏水呈细流状加入烧杯1中,边加边朝同一个方向搅拌,形成乳剂基质a,备用;
g)往乳剂基质a中加入凝胶A、凝胶B、凝胶C,形成基质b,备用;
h)将尿素、水杨酸、硼酸分散于1-6ml蒸馏水中,形成溶液a,备用;
i)将樟脑、冰片分散于1-5ml75%乙醇中,形成溶液b,备用;
j)往基质b中加入溶液a、溶液b、氮酮、丙三醇、1,2-丙二醇、茴香提取液、苦参提取液、5%尼泊金乙酯醇溶液,搅拌混合均匀,即得治疗灰指甲的乳剂。
