Abstract

Description

发明背景
已经建议将精氨酸和其它碱性氨基酸用于口腔保健,并且据信在对抗蛀洞(cavity)形成和牙敏感性方面具有显著益处。据信口腔中的碱性氨基酸是由某些种类的细菌代谢的,所述细菌为例如血链球菌(S.sanguis),这类细菌不产生龋齿并与致龋菌例如变性链球菌(S.mutans)竞争牙齿上和口腔中的位置。精氨酸分解细菌(arginolyticbacteria)可利用精氨酸和其它碱性氨基酸,产生氨气,从而升高其环境pH,而致龋菌则代谢糖,产生倾向于降低牙菌斑pH并使牙齿去矿化,最终导致蛀洞的乳酸。
目前用牙膏组合物(例如)来完成对口腔的精氨酸处理。也建议用精氨酸配制食品和饮品。尽管精氨酸在口腔中的有益作用在处理(例如刷牙、咀嚼香口胶或摄取富含精氨酸的食物)之后可维持数分钟或数小时,但是最好开发出将精氨酸递送到口腔的其它组合物和装置,例如与含有精氨酸的组合物联用或单独使用。
用于口腔的器具是本领域众所周知的,且包括例如牙刷、舌苔刮除器(tonguescrapers)、牙线、牙签(dentalpicks)、牙托(mouthguards)和牙齿矫正装置,例如牙套(braces)和固定器(retainers)。尽管这类器具可以用碱性氨基酸来处理,但碱性氨基酸可能会从这样的器具表面流失或消蚀掉,需要重复处理,而这可能会耗费时间而且对使用者来说也不方便。也可完成对这类器具的处理,而不完全或不正确的处理可能导致过少或过多碱性氨基酸掺入到器具表面。因此,迫切需要创建组合物、装置和方法,以克服这些问题。
发明概述
本发明涉及组合物和装置,所述组合物和装置将碱性氨基酸(例如精氨酸)递送到口腔,以便例如(i)减少或抑制龋齿的形成,(ii)减少、修复或抑制牙釉质的龋前缺损(pre-cariouslesions),例如如经定量光导荧光(QLF)或龋齿的电学测量(electricalcariesmeasurement,ECM)所检测的,(iii)减少或抑制牙齿去矿化并促进牙齿再矿化,(iv)降低牙超敏性,(v)减少或抑制牙龈炎,(vi)促进口腔内疮或伤口的愈合,(vii)降低产酸菌水平,(viii)提高精氨酸分解细菌的相对水平,(ix)抑制口腔内微生物生物膜的形成,(x)升高和/或维持牙菌斑pH水平为至少pH5.5,(xi)减少牙菌斑的积聚,(xii)清洁牙齿和口腔,(xiii)治疗口干症,(xiv)改善系统健康,和/或(xv)美白牙齿。这类组合物和装置可单独使用,或与其它含有碱性氨基酸的组合物(例如牙膏组合物)联用,以将碱性氨基酸递送到口腔。
本发明因此包括组合物1.0,一种包含包埋在生物可蚀性聚合物(bioerodablepolymer)中的碱性氨基酸的组合物。
本发明也包括下列组合物:
1.1.组合物1.0,其中所述碱性氨基酸是精氨酸、赖氨酸、瓜氨酸(citrullene)、鸟氨酸、肌氨酸、组氨酸、二氨基丁酸、二氨基丙酸、其盐和/或其组合。

2.将碱性氨基酸混合在所述溶液中;
3.将所述器具从所述溶液中取出,用所述溶液涂覆所述器具;和
4.蒸发所述溶剂。
优选地,所述器具不溶于或基本上不溶于所述溶剂。
本发明也包括制备用于口腔的器具的方法5,所述方法包括:
1.将碱性氨基酸与单体混合,所述单体可聚合形成生物可蚀性聚合物;
2.将器具浸泡在所述混合物中;
3.将所述器具从所述混合物中取出;和
4.在诱导单体聚合形成生物可蚀性聚合物的条件下处理所述清洁器具。
本发明还提供方法(方法6),包括达到以下目的的方法:(i)减少或抑制龋齿的形成,(ii)减少、修复或抑制牙釉质的龋前缺损,例如经定量光导荧光(QLF)或龋齿的电学测量(ECM)所检测的,(iii)减少或抑制牙齿去矿化并促进牙齿再矿化,(iv)降低牙超敏性,(v)减少或抑制牙龈炎,(vi)促进口腔内疮或伤口的愈合,(vii)降低产酸菌水平,(viii)提高精氨酸分解细菌的相对水平,(ix)抑制口腔内微生物生物膜的形成,(x)在糖攻击之后升高和/或维持牙菌斑pH水平为至少pH5.5,(xi)减少牙菌斑的积聚,(xii)治疗、减轻或减少口干症,(xiii)清洁牙齿和口腔,(xiv)减少侵蚀,(xv)美白牙齿,(xvi)使牙齿对致龋菌免疫;和/或(xvii)促进系统健康,包括心血管健康,例如通过降低经口腔组织所致的系统感染的可能性;

本发明还包括碱性氨基酸在制备牙科装置例如如上所述的装置3.0-3.9中的任一种的用途,例如用于上述制备方法例如方法4或5,或上述使用方法例如方法6。
本发明的其它实施方案对本领域技术人员来说将会是显而易见的。
发明详述
如本文所用的,生物可蚀性聚合物是可通过暴露于口腔而被降解的聚合物。当材料接触口腔中的物质例如唾液时可引起降解。也可因聚合物与口腔之间的摩擦和刺激而引起降解。根据聚合物的厚度,仅与口腔接触的聚合物表面被降解。
“有效量”是足以在口腔中具有想要的治疗效果或预防效果、而没有不适当的不良副作用例如刺激性或过敏反应的量。
除非另有说明,否则本文所用的碱性氨基酸包括呈游离碱和/或其盐的碱性氨基酸。
用于口腔的器具是本领域众所周知的,包括例如牙刷、舌苔刮除器、牙线、牙签、牙托和牙齿矫正装置,例如牙套和固定器。用于口腔的器具并不限于清洁和牙齿矫正装置。其它器具包括被设计用于口腔的物品,例如安抚奶嘴(pacifiers)(也称为橡皮奶头或抚慰器)和婴幼儿咀嚼玩具,例如牙胶环(teethingrings)。其它器具包括并非设计用于口腔的物品,但人们可以将这类物品放入他们的口中,例如钢笔和铅笔。
尽管这类器具可以用碱性氨基酸来处理,例如表面涂覆,但最好是将碱性氨基酸直接掺入到器具中,从而避免需要重复这类处理。这可通过以下方式来完成:通过用包含碱性氨基酸的生物可蚀性聚合物来涂覆器具,或用包含碱性氨基酸的生物可蚀性聚合物来制造器具。因为聚合物表面暴露在口腔内,所以聚合物表面的碱性氨基酸被释放到口腔中。因为聚合物表面被消蚀,例如在口腔内正常使用期间,所以额外的碱性氨基酸可被利用而不断释放到口腔中。
通过使用多孔材料,也可完成将碱性氨基酸直接掺入到器具中。器具可以由多孔材料来制备,或者涂覆多孔材料。通常,所述材料含有可存留碱性氨基酸的孔洞。当在例如口腔内使用器具时,氨基酸就被释放出来。如果孔洞释放其中所含的氨基酸,可以通过暴露于碱性氨基酸来处理多孔材料,以重新填充空洞。因此,碱性氨基酸可以从孔洞中释放出来或者捕获在孔洞中。
制备用于口腔的器具的方法、以及制备它们的组合物也都是本领域众所周知的。本发明也涵盖用组合物制备这类器具,所述组合物包含生物可蚀性聚合物和碱性氨基酸,或具有碱性氨基酸的多孔支架。
生物可蚀性聚合物是本领域技术人员众所周知的,且包括聚(乳酸)、聚(乙醇酸)和共聚物、聚二氧杂环己酮、聚(谷氨酸乙酯)、聚(羟基丁酸酯)、聚羟基戊酸酯和共聚物、聚己内酯、聚酐、聚(原酸酯)、聚(亚氨基碳酸酯)、聚酯酰胺、聚酯胺、聚氰基丙烯酸酯、聚磷腈、共聚物和其它脂族聚酯或其合适的共聚物包括聚-L-乳酸和聚-e-己内酯的共聚物;混合物、共聚物及其组合。其它有用的聚合物也可包括聚丙烯酸酯、聚甲基丙烯酸酯、聚脲、聚氨酯、聚烯烃、聚卤代乙烯(polyvinylhalides)、聚偏二卤乙烯(polyvinylidenehalides)、聚乙烯醚、polyvinalaromatics、聚乙烯酯、聚丙烯腈(polyactylonitriles)、树脂、聚硅氧烷、环氧树脂和聚四氟乙烯。其它有用的聚合物可包括或基于血纤蛋白、胶原蛋白、糖胺聚糖、低聚糖和多糖、软骨素、磷脂、磷酸胆碱、脱乙酰壳多糖、藻酸盐、纤维(fibring)、血纤蛋白原、纤维素塑料、淀粉、葡聚糖、糊精、透明质酸、肝素和弹性蛋白。聚合物也可以是例如由乙交酯、丙交酯、E-己内酯和聚乙二醇的基本单元构建的多嵌段共聚物。有用的聚合物也包括聚(酯-酰胺)聚合物或同系物,其基于亮氨酸、苯丙氨酸和/或精氨酸,任选具有一种或多种二醇和一种或多种二羧酸。
自例如单体形成这类聚合物的方法是本领域众所周知的。诱导聚合的方法是本领域已知的,例如溶于溶剂中,暴露给辐射,包括可见光和紫外辐射和热处理。
多孔支架是本领域已知的,且包括塑料、陶瓷和其它材料。塑料及其形成是本领域众所周知的,并包括聚乙烯、聚甲基丙烯酸甲酯、聚氨酯、聚对苯二甲酸乙二醇酯、聚丙烯、聚苯乙烯、聚酰胺、生物塑料和生物可降解塑料。孔洞的尺寸很重要,因为小的孔洞尺寸不能稳定吸收和释放碱性氨基酸,而大的孔洞将需要稳定的负载。在本发明的一个实施方案中,孔洞的平均直径为约10μm至约500μm,例如约10μm、20μm、30μm、40μm、50μm、60μm、70μm、80μm、90μm、100μm、200μm、300μm或400μm直径。
可用于本发明组合物的碱性氨基酸不仅包括天然发生的碱性氨基酸例如精氨酸、赖氨酸和组氨酸,而且包括分子中具有羧基和氨基的任何碱性氨基酸。因此,碱性氨基酸包括但不限于精氨酸、赖氨酸、瓜氨酸、鸟氨酸、肌氨酸、组氨酸、二氨基丁酸、二氨基丙酸、其盐或其组合。在一个具体的实施方案中,碱性氨基酸选自精氨酸、瓜氨酸和鸟氨酸,优选精氨酸,例如1-精氨酸。
可在聚合之前或期间,将碱性氨基酸以游离碱或盐形式添加到生物可蚀性聚合物中。如本文所用的,提及碱性氨基酸时也包括其盐。因为本发明的组合物和装置预期用于口腔,用于本发明的盐在所提供的用量和浓度下,对这类使用而言应当是安全的。合适的盐包括本领域已知的为药学上可接受的盐的那些盐,通常认为在所提供的用量和浓度下是生理上可接受的。生理上可接受的盐包括由药学上可接受的无机或有机酸或碱所衍生的那些,例如由形成生理上可接受的阴离子的酸所形成的酸加成盐(例如盐酸盐和氢溴酸盐),和由形成生理上可接受的阳离子的碱所形成的碱加成盐,例如衍生自碱金属(例如钾和钠)的那些或衍生自碱土金属(例如钙和镁)的那些。可以用本领域已知的标准方法来获取生理上可接受的盐,所述方法是例如通过使足够碱性的化合物例如胺与合适的酸反应,生成生理上可接受的阴离子。
本发明的组合物包含有效量的碱性氨基酸。因此,所述组合物可包含约1%至约50%重量的碱性氨基酸,例如约1、2、3、4、5、10、15、20、25、30、35、40或45%重量。
在一个实施方案中,所述碱性氨基酸可溶于聚合物。在一个优选的实施方案中,所述碱性氨基酸不溶于聚合物。如果氨基酸不溶,优选氨基酸的粒径足够小,从而当用于口腔时不会引起对口腔的刺激。优选地,所述氨基酸在聚合物中均匀地分布。
在一个实施方案中,所述聚合物是阴离子型聚合物,例如聚羧酸酯聚合物,例如丙烯酸酯聚合物或共聚物,其中碱性氨基酸与聚合物中的羧酸部分形成盐。
在本发明的一个实施方案中,生物可蚀性聚合物或多孔支架可任选包括有效量的氟化物或氟离子源。多种多样的产生氟离子的原料都可用作本发明组合物中的可溶性氟化物的来源。合适的产生氟离子的原料的实例可参见Briner等的美国专利号3,535,421;Parran,Jr.等的美国专利号4,885,155和Widder等的美国专利号3,678,154,这些专利文献通过引用结合到本文中。代表性的氟离子源包括但不限于氟化亚锡、氟化钠、氟化钾、一氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺、氟化铵,及其组合。在某些实施方案中,氟离子源包括氟化亚锡、氟化钠、一氟磷酸钠及其混合物。
在本发明的另一实施方案中,生物可蚀性聚合物或多孔支架可任选包含选自以下的防腐剂或抗微生物剂:三氯生、草药提取物和精油(例如迷迭香提取物、麝香草酚、薄荷醇、桉油醇、水杨酸甲酯)、双胍防腐剂(例如氯己定、阿来西定或奥替尼啶)、季铵化合物(例如氯化十六烷基吡啶)、酚类防腐剂、海克替啶、聚维酮碘、地莫匹醇、5-正辛酰-3’-三氟甲基苯基水杨酰胺(salifluor)、金属离子(例如锌盐,例如柠檬酸锌)、血根碱、蜂胶,及其组合,以便进一步增强碱性氨基酸的有益效果。
当涂覆在器具上时,优选生物聚合物或多孔支架足够薄而不会妨碍器具的使用,但又足够厚而有足够量的聚合物持续涂覆在器具上用于随后的正常使用,例如使用3、4、5、10、15、20、25、30、60或90次。器具的正常使用将取决于具体器具。生物聚合物的厚度可以是约0.1μm至约3000μm,约1μm、5μm、10μm、25μm、50μm、100μm、250μm、500μm、1000μm、2000μm或约3000μm厚。涂覆的厚度将取决于具体器具。
本发明的组合物也可包括矫味剂。本发明实践中所用的矫味剂包括但不限于精油以及各种矫味用的醛、酯、醇和类似原料。精油的实例包括留兰香油、薄荷油、冬青油、黄樟油、丁香油、鼠尾草油、桉树油、甘牛至油、肉桂油、柠檬油、白柠檬油、葡萄柚油和甜橙油。也可使用诸如薄荷醇、香芹酮和茴香脑的此类化学物。本发明的组合物也可包括着色剂。矫味剂和着色剂的使用可有助于确定支架是否含有氨基酸,或生物可蚀性聚合物的降解程度。
本发明的组合物也可用作口腔内的粘合剂,例如用于支架(brackets)与矫形牙套(braces)的粘合。本发明的组合物也可用作器具生产的粘合剂,例如将牙刷头与柄粘合。本发明也可用作牙科密封剂,例如保护性涂层。
实施例1
用包含40%重量L-精氨酸和矫味剂例如薄荷的生物可蚀性聚合物涂覆适用于接触性运动的洁净牙托。使用期间,聚合物表面被降解,将L-精氨酸释放到例如口腔中,以减少或抑制龋齿的形成,减少、修复或抑制牙釉质的龋前缺损,减少或抑制牙齿去矿化并促进牙齿再矿化,减少或抑制牙龈炎,降低产酸菌水平,提高精氨酸分解细菌的相对水平并升高和/或维持牙菌斑pH水平为至少pH5.5。重复使用该牙托进一步降解聚合物,当牙托不再有薄荷味时,使用者知道聚合物已完全降解掉。
实施例2
用平均孔径为50um的多孔塑料支架制造安抚奶嘴。将安抚奶嘴浸泡在包含精氨酸碳酸氢盐的洁净溶液中,使精氨酸碳酸氢盐吸附在孔洞中。
实施例3
将实施例2的安抚奶嘴给予婴儿,进行正常使用,即吮吸和咀嚼。当婴儿吮吸和咀嚼安抚奶嘴时,精氨酸碳酸氢盐就释放到例如口腔中,以抑制口腔中微生物生物膜的形成。
实施例4
将实施例3中使用过的安抚奶嘴浸泡在包含精氨酸碳酸氢盐的洁净溶液中。
实施例5
用包含精氨酸和着色剂例如红色色素的生物可蚀性聚合物涂覆洁净牙刷头。刷毛附着在头部,而头部连接柄。在口腔中使用牙刷时,随着聚合物被降解,精氨酸会被释放出来。当头部不呈红色时,使用者知道头部或牙刷需要更换。
Claims (32)
1.一种器具,选自牙刷、牙签、舌头清洁器、护口器和牙齿矫正装置,其中所述器具的至少一部分涂覆有组合物,该组合物包含包埋在生物可蚀性聚合物中的呈游离或盐形式的碱性氨基酸。
2.权利要求1的器具,其中所述生物可蚀性聚合物选自聚(乳酸)、聚(乙醇酸)和共聚物;聚二氧杂环己酮、聚(谷氨酸乙酯)、聚(羟基丁酸酯)、聚羟基戊酸酯和共聚物、聚己内酯、聚酐、聚(原酸酯)、聚(亚氨基碳酸酯)、聚酯酰胺、聚酯胺、聚氰基丙烯酸酯、聚磷腈、共聚物和其它脂族聚酯;或其合适的共聚物,包括聚-L-乳酸和聚-e-己内酯的共聚物;血纤蛋白、胶原蛋白、糖胺聚糖、低聚糖、多糖、软骨素、脱乙酰壳多糖、藻酸盐、纤维、血纤蛋白原、纤维素塑料、淀粉、葡聚糖、糊精、透明质酸、肝素、弹性蛋白;由乙交酯、丙交酯、E-己内酯和聚乙二醇的基本单元构建的多嵌段共聚物;和聚(酯-酰胺)聚合物或同系物,其基于亮氨酸、苯丙氨酸和/或精氨酸,任选具有一种或多种二醇和一种或多种二羧酸。
3.一种组合物,其包含具有多孔的支架和存在于所述孔内的碱性氨基酸。
4.权利要求3的组合物,其中所述支架包含塑料。
5.权利要求3的组合物,其中所述支架包含选自聚乙烯、聚甲基丙烯酸甲酯、聚氨酯、聚对苯二甲酸乙二醇酯、聚丙烯、聚苯乙烯、聚酰胺、生物塑料、生物可降解塑料及其组合的材料。
6.权利要求3的组合物,其中所述碱性氨基酸可逆地存在于所述孔内。
7.权利要求1的器具,其中所述碱性氨基酸选自精氨酸、赖氨酸、瓜氨酸、鸟氨酸、肌氨酸、组氨酸、二氨基丁酸、二氨基丙酸、其盐和/或其组合。
8.权利要求1的器具,其中所述碱性氨基酸具有L-构型。
9.权利要求1的器具,其中所述碱性氨基酸是精氨酸。
10.权利要求8的器具,其中所述碱性氨基酸是L-精氨酸。
11.权利要求1的器具,其中所述碱性氨基酸呈盐形式。
12.权利要求11的器具,其中所述碱性氨基酸的盐形式是精氨酸磷酸盐。
13.权利要求11的器具,其中所述碱性氨基酸的盐形式是精氨酸盐酸盐。
14.权利要求1的器具,还包含阴离子型聚合物。
15.权利要求11的器具,其中所述碱性氨基酸的盐形式是精氨酸碳酸氢盐。
16.权利要求1的器具,其中所述器具是包括头部的牙刷,所述头部上附有刷毛。
17.权利要求16的器具,其中所述刷毛被涂覆。
18.权利要求16的器具,其中所述头部被涂覆。
19.权利要求1的器具,其中所述器具是牙签。

27.权利要求1的器具,其中所述组合物还包含钾离子源。
28.权利要求27的器具,其中所述钾离子源选自硝酸钾和氯化钾。
29.权利要求3的组合物,其中所述组合物还包含氟化物离子源。
30.权利要求29的组合物,其中所述氟化物离子源选自氟化亚锡、氟化钠、氟化钾、一氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺、氟化铵及其组合。
31.权利要求3的组合物,其中所述组合物还包含钾离子源。
32.权利要求31的组合物,其中所述钾离子源选自硝酸钾和氯化钾。
