Abstract

L'invention concerne des procédés et des compositions pour le coiffage d'ARN dans un mélange de transcription in vitro (TIV) qui comprennent un variant d'ARN polymérase thermostable et un analogue de coiffe, de sorte que, lorsqu'un modèle d'ADN est ajouté au mélange et que le mélange est ensuite incubé dans des conditions de transcription in vitro , un ARN coiffé est produit. Les procédés décrivent des rendements de coiffage élevés à des températures élevées ainsi qu'à des températures standard pour la TIV. L'ARN polymérase utilisé est un variant thermostable de l'ARN polymérase T7, appelé polymérase M20, qui comprend des substitutions aux positions 388 et 567, notamment D388E et V567P.
