Abstract

L'invention concerne une méthode servant à immobiliser un nucléotide ou un polynucléotide sur un support solide, consistant à : fournir un support solide présentant une surface fonctionnalisée de formule B - S - S L, dans laquelle B est le support solide et L est un groupe partant ; fournir un nucléotide ou un polynucléotide comprenant au moins un groupe phosphorothioate ; et coupler les deux ensemble grâce à une réaction d'échange sulfurée. Le produit présente la structure B - S - S - P(O)R?1¿R?2¿, dans laquelle - POR?1¿R?2¿ représente un nucléotide ou un polynucléotide. Les ensembles de polynucléotides immobilisés sont utiles pour la mise en oeuvre de techniques APEX ou SBH.
