Arquéias são os mais antigos microorganismos existentes na natureza, porém, somente foram descritas recentemente (Woese CR & Fox GE. Proc Natl Acad Sci USA 74:5088-5090,1977). São caracterizadas por habitarem em locais que usualmente não permitem a vida, como águas extremamente salgadas, soluções com alta temperatura e em solventes. Recentemente, um novo tipo de archaea, extremamente pequena, dependente das archaeas maiores, foi descrito e denominado de nanoarchaea (Huber J e cols. Nature 417:63-67,2002). No documento de patente anterior BR PI0300914-9 foram descritas arquéias patogênicas em lesões do tipo calcificação de valva aórtica, neoplasias e fíbrose hepática esquistossomótica, tendo sido, em trabalhos posteriores, apresentados maiores detalhamentos das arquéias patogênicas em placas de ateroma vulneráveis (Higuchi e cols. Clinics 61:473-478, 2006), e na cardiopatia crônica chagásica (Higuchi e cols. Não obstante aos avanços nas técnicas de detecção de placas de aterosclerose, necessita-se ainda encontrar marcadores mais precisos de doença aterosclerótica coronariana (DAC) e de instabilidade da placa. Em trabalhos prévios, detectou-se a presença de antígenos de Mycoplasma pneumoniae (MP) e Proteína C Reativa (PCR) nas placas ateroscleróticas e, nas vulneráveis a presença de arquéias patogênicas. Aqui investigou-se se esses elementos podem ser detectados no soro de pacientes com DAC para utilizá-los como biomarcadores. Material e Métodos: Dois ml de soro de idosos, 13 indivíduos saudáveis e 12 com DAC, foram analisadas por imuno-microscopia eletrônica (ME) para detecção de partículas com antígenos de MP ou PCR, marcadas com ouro coloidal. Foram feitas 5 fotomicrografias/5.500x e obtidos os números médios de partículas positivas4im2.Despite advances in atherosclerosis plaque detection techniques, more accurate markers of coronary atherosclerotic disease (CAD) and plaque instability still need to be found. MATERIALS AND METHODS: Two ml of serum from the elderly, 13 healthy individuals and 12 with CAD, were analyzed by electron microscopy (EM) to detect particles with colloidal gold-labeled PM or PCR antigens. Five photomicrographs / 5,500x were made and the average numbers of positive particles 4im2 were obtained.
