Abstract
MéTODO PARA A PRODUçãO EM MASSA DE LECTINA LIGANTE DE MANOSE MULTIMéRICA. Trata-se de um método paira a produção em massa de lectina ligante de manose humana recombinante multimérica (rhMBL) em uma linhagem de células CHO transfectada com um vetor contendo a seqüência da região codificadora de MBL humana. Ainda, esta invenção refere-se à construção de uma linhagem de células recombinantes da qual MBL pode ser produzida mais que 80% como forma polimérica altamente funcional. Ainda, essa invenção refere-se à construção de uma linhagem de células recombinantes que pode produzir 50 <109>g/milhão de células/dia em um sistema de cultura em frasco para a produção de MBL. Ainda, esta invenção descreve métodos para cultura de alta densidade da célula em um meio isento de proteína usando biorreator e para a purificação seletiva da forma de alto peso molecular da MBL do meio de cultura. Essa invenção descreve também um método para a produção de rhMBL funcional que pode ser usada no desenvolvimento de um agente terapêutico para o tratamento de infecções virais, bacterianas, ou fúngicas.
