"MéTODO PARA DIAGNOSTICAR A GRAVIDADE DE UMA DOENçA EM UM INDIVìDUO DE TESTE, E, KIT DE TESTE PARA TRIAGEM DE UMA AMOSTRA DE PACIENTE QUANTO à PRESENçA DE CéLULAS CANCEROSAS CIRCULANTES" Um ensaio altamente sensível é apresentado, o qual utiliza um método para ampliação preparada específica de genes, de coleções de MRNA de células raras e de transcritos raros encontrados no sangue. O ensaio possibilita a detecção de MRNA raro (10 cópias/célula) encontrado em 1 a 10 células isoladas através de enriquecimento imunomagnético. O ensaio é um melhoramento sobre a PCR multicomplexa e permite a detecção eficiente de seqüências codificadores raras para células de carcinomas circulantes no sangue. Os métodos são úteis no perfilamento de células isoladas de tecidos ou de fluidos corporais e servem como um adjunto para o diagnóstico clinico de diversos carcinomas, incluindo a detecção de estágio antecipada e a classificação de células tumorais circulantes. The assay enables detection of rare MRNA (10 copies / cell) found in 1 to 10 isolated cells by immunomagnetic enrichment. The assay is an improvement over multicomplex PCR and allows efficient detection of rare coding sequences for circulating carcinoma cells in the blood. The methods are useful in profiling cells isolated from tissues or body fluids and serve as an adjunct to the clinical diagnosis of various carcinomas, including early stage detection and classification of circulating tumor cells. Monitoring of nucleic acid and protein profiles of cells, either in conventional or microarray formats, facilitates control of therapeutic intervention, including staging, monitoring response to therapy, confirmation of remission, and detection of regression.
