Abstract

A method for the detection of cytosine methylations in genomic DNA is disclosed, whereby genomic DNA samples are firstly reacted with a chemical reagent, whereupon 5-methylcytosine and cytosine react differently and thus display a different base pair relationship in the DNA duplex after said reaction. The pre-treated DNA is then amplified by means of a polymerase and at least one primer oligonucleotide and the amplified material once again reacted with a chemical reagent as above. The cytosine bases and/or guanine bases remaining in the amplified material after the renewed chemical treatment are detected in the final step.
