Abstract

Un método para la detección directa de mecanismos de resistencia mediante Espectrometría de masas MALDI-TOF. El método comprende preparar diferentes cultivos bacterianos y además utilizar una técnica de ácido sinapínico para la detección de proteína intacta, en donde cada cultivo bacteriano está contenido en una mancha objetivo MALDI. El método también comprende adquirir un espectro de cada cultivo estableciendo el rango de espectro de un espectrómetro de masas, usando un programa de computación para MALDI-TOF, a un rango comprendido entre 17.000 y 50.000 Da. Finalmente, se comparan los diferentes espectros adquiridos teniendo en cuenta un peso teórico de la proteína a detectar.A method for the direct detection of resistance mechanisms by MALDI-TOF mass spectrometry. The method comprises preparing different bacterial cultures and also using a sinapinic acid technique for the detection of intact protein, where each bacterial culture is contained in a MALDI target spot. The method also comprises acquiring a spectrum of each culture by setting the spectrum range of a mass spectrometer, using a computer program for MALDI-TOF, to a range between 17,000 and 50,000 Da. Finally, the different acquired spectra are compared taking into account a theoretical weight of the protein to be detected.
