Abstract
Cebadores, sondas y conjuntos de elementos (kits) para la deteccion de secuencias variantes de EML4-ALK en una muestra y métodos de deteccion de variantes de EML4-ALK usando los cebadores, sondas y conjuntos de elementos que se divulgan. Reivindicacion 1: Un método para detectar la presencia de una variante EML4-ALK en una muestra, caracterizado porque dicho método comprende: (a) aislar un ácido nucleico de dicha muestra, en donde la muestra comprende secuencias de ADN; (b) conducir una reaccion de amplificacion de dichas secuencias de ADN de dicha muestra, en donde dicha reaccion de amplificacion comprende un primer cebador capaz de alinearse específicamente con una secuencia de una variante EML4-ALK en una primera posicion en una secuencia de ADN de EML4-ALK, en donde dicho primer cebador se selecciona del grupo que consiste de la SEQ ID N°:1, la SEQ ID N°:2, la SEQ ID N°:3, la SEQ ID N°:4, la SEQ ID N°:5, la SEQ ID N°:6, la SEQ ID N°:7, la SEQ ID N°:8, la SEQ ID N°:9, la SEQ ID N°:12, la SEQ ID N°:13, la SEQ ID N°:14, la SEQ ID N°:15, la SEQ ID N°:16, la SEQ ID N°:17, la SEQ ID N°:18, la SEQ ID N°:19, la SEQ ID N°:20, la SEQ ID N°:21, la SEQ ID N°:22, la SEQ ID N°:23, la SEQ ID N°:24 y la SEQ ID N°:25, y un segundo cebador capaz de alinearse específicamente en una segunda posicion en una secuencia de ADN de EML4-ALK, en donde dichos primer y segundo cebadores se alinean con diferentes cadenas de secuencias de ADN de cadena doble de EML4-ALK, en donde la reaccion de amplificacion tiene la capacidad de producir un producto de amplificacion específico de la variante EML4-ALK cuando las secuencias de ADN de la muestra comprenden una secuencia de ADN de EML4-ALK que comprende una secuencia de una variante en dicha primera posicion de dicha secuencia de ADN de EML4-ALK; y (c) visualizar los productos de amplificacion producidos por dicha reaccion de amplificacion, en donde la deteccion de un producto de amplificacion específico de la variante EML4-ALK constituye un indicador positivo de una variante EML4-ALK en dicha muestra.Primers, probes and element sets (kits) for the detection of EML4-ALK variant sequences in a sample and methods of detection of EML4-ALK variants using the primers, probes and element sets disclosed. Claim 1: A method for detecting the presence of an EML4-ALK variant in a sample, characterized in that said method comprises: (a) isolating a nucleic acid from said sample, wherein the sample comprises DNA sequences; (b) conducting an amplification reaction of said DNA sequences of said sample, wherein said amplification reaction comprises a first primer capable of specifically aligning with a sequence of an EML4-ALK variant in a first position in a DNA sequence of EML4-ALK, wherein said first primer is selected from the group consisting of SEQ ID N °: 1, SEQ ID N °: 2, SEQ ID N °: 3, SEQ ID N °: 4, SEQ ID N °: 5, SEQ ID N °: 6, SEQ ID N °: 7, SEQ ID N °: 8, SEQ ID N °: 9, SEQ ID N °: 12, SEQ ID N °: 13, SEQ ID N °: 14, SEQ ID N °: 15, SEQ ID N °: 16, SEQ ID N °: 17, SEQ ID N °: 18, SEQ ID N °: 19, SEQ ID N °: 20, SEQ ID N °: 21, SEQ ID N °: 22, SEQ ID N °: 23, SEQ ID N °: 24 and SEQ ID N °: 25, and a second primer capable of specifically aligning in a second position in an EML4-ALK DNA sequence, wherein said first and second primers align with different strands of chain DNA sequences. double of EML4-ALK, where the amplification reaction has the ability to produce a specific amplification product of the EML4-ALK variant when the DNA sequences of the sample comprise an EML4-ALK DNA sequence comprising a sequence of a variant in said first position of said EML4-ALK DNA sequence; and (c) visualizing the amplification products produced by said amplification reaction, wherein the detection of a specific amplification product of the EML4-ALK variant constitutes a positive indicator of an EML4-ALK variant in said sample.
