Abstract
Esta solicitud revela un novedoso proceso para la preparacion de 2-hidroxi-N,N-dimetil-3-[[2-[[1(R)-(5-metil-2-furanil)propil]amino]-3,4-dioxo-1-ciclobuten-1-il]amino]benzamida, que tiene utilidad, por ejemplo, en el tratamiento de enfermedades mediadas por CXC quimioquinas. Reivindicacion 1: Un proceso para proporcionar una masa cristalina de la Forma 4 monohidrato del compuesto de formula (1) que proporciona una resistencia específica de torta de filtracion de menos de 7,9 X 10 11 m/kg, segun se mide de acuerdo con el Procedimiento de Medicion de la Resistencia Específica de Torta ôV(i)ö descrito en la presente, (1) donde el proceso comprende: (a) proporcionar una solucion del compuesto de formula (1) en una mezcla de solvente y anti-solvente a la temperatura de disolucion del compuesto de formula (1) proporcionado por el sistema de solvente/anti-solvente seleccionado; (b) enfriar la solucion del paso ôaö hasta una temperatura justo por encima de una temperatura a la cual comienza la nucleacion del compuesto de formula (1) en la mezcla de solvente/anti-solvente seleccionada para el paso ôaö y sembrar el lote con una forma cristalina solida del compuesto de formula (1) Forma 4 monohidrato, con lo cual se forma una mezcla; (c) enfriar la mezcla del paso ôbö hasta una temperatura a la cual sustancialmente todo el compuesto de formula (1) disuelto en el paso ôaö se cristaliza en una suspension, usando una velocidad de enfriamiento de entre aproximadamente 0,01°C/minuto y aproximadamente 5°C/minuto; y (d) ciclando la temperatura de la suspension del paso ôcö calentándola hasta una temperatura por debajo de la temperatura de siembra empleada en el paso ôbö, a una velocidad de entre aproximadamente 0,01°C/minuto hasta aproximadamente 5°C/minuto, y enfriando la suspension calentada a una velocidad de entre aproximadamente 0,01°C/minuto y aproximadamente 5°C/minuto, hasta una temperatura de aproximadamente la temperatura de cristalizacion alcanzada en el paso ôcö y repitiendo esto hasta obtener cristales de la seccion transversal deseada, lo cual proporciona una resistencia específica de torta de filtracion de menos de 7,9 X 10 11 m/kg cuando los cristales precipitados se aíslan por filtracion.
