Abstract

Foreliggende oppfinnelse tilveiebringer en fremgangsmåte for å fremstille et polypeptid som har en C-terminal a-karboksamidgruppe. Spesielt omhandlet er en enzymatisk modifisering av valgte substratpolypeptider som resulterer i kløyving av substrarpolypeptidet for å danne et produktpeptid med en C-terminal argininrest, som har en a- karboksamidgruppe (C-terminal «Arg-NH2»). Fremgangsmåten inkluderer å kontakte en vannbasert løsning som inkluderer (i) ammoniakkreagens og (ii) substratpolypeptidet med (iii) klostripain.
