Abstract

La invención se refiere a un método para preparar vesículas con gradiente de pH de transmembrana; este método se caracteriza por los siguientes pasos: a) preparar vesículas hechas de por lo menos una sustancia de matriz en un medio acuoso que tiene una osmolaridad de no más de 200 mOsmoles/l, en donde la sustancia de matriz se selecciona del grupo que consiste de lípidos anfifílicos y copolímeros de bloque anfifílicos, b) transferir las vesículas en un regulador de pH básico o ácido que tiene una osmolaridad que es por lo menos 200 mOsmoles/l mayor que la osmolaridad del medio acuoso del paso a) para aplicar un choque osmótico a las vesículas y para obtener vesículas llenas de regulador de pH, y c) diluir una mezcla del medio acuoso y el regulador de pH básico o ácido que contiene a las vesículas llenas de regulador de pH añadiendo una solución neutralizante para obtener vesículas con gradiente de pH de transmembrana suspendidas en un regulador de pH de suspensión, en donde el regulador de pH de suspensión difiere del regulador de pH básico o ácido en el valor del pH.
