Abstract
La presente invención se refiere a un procedimiento para la expresión periplásmica de un toxoide bacteriano, que comprende los pasos de: (a) desarrollar un cultivo de una célula hospedera gram negativa en un medio de fermentación, en donde la célula hospedera es transformada con un polinucleótido, y en donde el polinucleótido codifica el toxoide bacteriano y una secuencia de señal periplásmica; o proveer una célula hospedera gram negativa en donde la célula hospedera es transformada con un polinucleótido, el polinucleótido codifica el toxoide bacteriano y una secuencia de señal periplásmica, y en donde la célula hospedera gram negativa comprende el toxoide bacteriano expresado en el periplasma; (a)(i) inducir la expresión del toxoide bacteriano; (b) hacer madurar la célula hospedera, en donde el paso de maduración comprende: (I) someter la célula hospedera a un choque de pH; (II) incubar la célula hospedera sin la adición de alimento; y/o (III) someter la célula hospedera a una temperatura por abajo de -20°C; y (c) extraer el toxoide bacteriano de la célula hospedera, en donde el procedimiento de extracción comprende un choque osmótico.
