Abstract
La presente descripción proporciona sistemas, métodos y composiciones para la ingeniería genética específica del sitio usando la adición programable a través de elementos objetivo específicos del sitio (PASTE). PASTE comprende la adición de un sitio de integración en un genoma objetivo seguida por la inserción de uno o más genes de interés o una o más secuencias de ácidos nucleicos de interés en el sitio. PASTE combina las tecnologías de edición de genes y las tecnologías de integrasa para lograr la incorporación unidireccional de genes en un genoma para el tratamiento y diagnóstico de enfermedades.
