Abstract
La mucine 16 (muc16) est fortement exprimée dans le cancer de l'ovaire et son expression sur des cellules cancéreuses est démontrée pour protéger des cellules tumorales du système immunitaire. La présente invention concerne de nouveaux anticorps igg humains de pleine longueur qui se lient à muc16 humaine (anticorps monospécifiques). La présente invention concerne également de nouveaux anticorps bispécifiques (bsabs) qui se lient à la fois à muc16 et à cd3 et activent les lymphocytes t par l'intermédiaire du complexe cd3 en présence de tumeurs exprimant muc16. Selon certains modes de réalisation, la présente invention concerne des molécules de liaison aux antigènes bispécifiques comprenant un premier domaine de liaison aux antigènes qui se lie spécifiquement au cd3 humain et du singe, et une seconde molécule de liaison aux antigènes qui se lie spécifiquement à muc16 humaine et du singe. Dans certains modes de réalisation, les molécules de liaison aux antigènes bispécifiques selon la présente invention sont capables d'inhiber la croissance de tumeurs exprimant muc16. The present invention relates to novel full-length human igg antibodies that bind to human muc16 (monospecific antibodies). The present invention also relates to novel bispecific antibodies (bsabs) that bind to both muc16 and cd3 and activate t cells through the cd3 complex in the presence of muc16-expressing tumors. Embodiments of the present invention provide bispecific antigen-binding molecules comprising a first antigen-binding domain that specifically binds to human and monkey cd3, and a second antigen-binding molecule that specifically binds to muc16 human and monkey. In certain embodiments, the bispecific antigen-binding molecules of the present invention are capable of inhibiting the growth of muc16-expressing tumors. The bispecific antigen-binding molecules of the invention are useful for the treatment of diseases and disorders in which an induced or upregulated muc16-targeting immune response is desired and/or therapeutically beneficial. For example, the bispecific antibodies of the invention are useful for the treatment of various cancers, including ovarian cancer. The present invention also relates to drug-anti-muc16 antibody conjugates that inhibit tumor growth in vivo. In some embodiments, the anti-muc16 antibodies are useful in diagnostic methods to identify the presence of muc16 in tissue and/or plasma samples.
