Abstract
Det beskrives en fremgangsmåte for frigjøring av et nukleinsyremolekyl fra en fast bærer som det er festet til, hvor et ukonvensjonelt nukleotid er inkorporert i et forutbestemt sete i nevnte nukleinsyremolekyl, hvor fremgangsmåten omfatter selektiv avspaltning av nevnte nukleinsyremolekyl fra setet for nevnte ukonvensjonelle nukleotid. Nukleinsyremolekylet kan være et kimært molekyl som omfatter en nukleinsyre (nukleotidsekvens) og en annen molekylkomponent med en forskjellig kjemisk natur. Derved tilveiebringes også et kimært molekyl (eller konstruksjon) som omfatter en nukleotid-linkersekvens omfattende et selektivt avspaltbart ukonvensjonelt nukleotid fra et forutbestemt sete, koblet til en funksjonell gruppe, som kan være en affinitetsbindingsgruppe eller en rapportørgruppe. Anvendelsesområder kan for eksempel være molekylærbiologiske prosedyrer, så som nukleinsyresyntese, amplifikasjon eller sekvensering, nukleinsyreisolering eller affinitets-baserte separasjons- eller testprosedyrer.
