the lens holder: when you insert the lens, does it scratch the surface of the lens? and in your design, how to fix the holder on stereotax?
and how to remove the holder after implating the lens?
Hi @ChangQing-SH,
This is more or less our protocol. In the future we will add some images to clarify.
We fix the lens holder to a lift table lab stand (similar to this one https://www.amazon.com/dp/B07L834W1J), with tape in both side wings, over a calibration slide. Then we manually insert the lens. Just being careful, we have done this several times without scratching the lens surface.
We put the miniscope into the miniscope-holder and then the baseplate on the miniscope.
We lower the miniscope with the baseplate onto the lens until image from slide comes into focus.
We glue the lens to baseplate just using a bit of superglue, then left it rest until its dry. Gel superglue is easier but takes longer to dry. Liquid superglue dries quicker but it can happen that, because of a capillarity effect, the superglue will cover the surface of the lens. if this happens, you will notice that the image that was in focus, suddenly gets blurred. We just use the gel superglue.
Once dry, loose the baseplate screw from the miniscope, carefully lift the miniscope. The baseplate will remain attached to the lens, so they will either stay in the lens holder or they will come up with the miniscope but either way is easy to detach the baseplate from the miniscope.
Under a microscope inspect the union between the lens and the baseplate and add more superglue if necessary.
Hi @ChangQing-SH <https://github.com/ChangQing-SH>,
We don't actually have a surgical protocol specifically written down. We
base our procedures on those described at miniscope.org, which has
extensive video and photo documentation of the surgical process.
For coating the GRIN lens with silk/virus, see:
https://pubmed.ncbi.nlm.nih.gov/29562189/
Good luck!

thanks,
it's great to see this with some images!
Btw, I am also a newbie to the Miniscope, Can you also share a detailed V4 surgical protocol with me?
best,
